With continuing depletion of petroleum reserves and releasing of enormous greenhouse gas,the development and utilization of renewable energy has caught wide attention.Conversion from biomass to platform chemicals and bulk chemicals is an important developing orientation of biomass conversion.Due to the similar structure,2,5-Furandicarboxylic acid(FDCA)can replace the terephthalicacid in the production of polyethylene terephthalate(PET).FDCA can be produced by oxidation of 5-hydroxymethylfurfural(HMF).In contrast to chemical synthesis,biocatalytic transformation is a better alternative route which is typically performed under relative mild conditions.In these process,HMFO can oxidize HMF to 2,5-diformylfuran(DFF)and 5-formylfuroic acid(FFA)without the addition of FAD cofactor.Subsequently,lipase oxidizes DFF and FFA to FDCAcompletely.Enzymatic cascade oxidation of HMF to FDCA was performed with a yield of 94.01% by combination of 5-hydroxymethylfurfural oxidase(HMFO)and lipase.The FDCA purity was improved from 84.43% to 99% with recovery of 78.10%by employing alkali precipitation in organic solvent,which is coupled with second step reaction.In addition,coming over the oxidation of 5-formylfuroic acid by site-directed mutagenesis of 5-hydroxymethylfurfural oxidase.The conversion from 5-hydroxymethylfurfural to 2,5-Furandicarboxylic acidwas realized by combination of 5-hydroxymethylfurfural and mutagenic 5-hydroxymethylfurfural in one-pot reaction,and led to a2,5-furandicarboxylic acid yield of 97.17%,superiorto the method of dual enzyme cascade reaction which yield is 60.88%.
