In order to solve the traditional problems of labor,long waiting time,the detection method of the microfluidic chip and surface enhanced Raman combination,complete the analysis of the operation process of the sample reaction,based on a microfluidic chip.Choose the nano silver particles and gold nano particle as a substrate for Raman detection,because the metal nano materials have unique optical properties of noble metal nanoparticles,specific size can produce a "hot spot" effect,the SERS enhancement factor can reach a maximum of 107~1014 times,so as to realize the detection of high sensitivity.The specific contents of this paper are as follows:(1)The microfluidic chip was used to detect the content of R6 G and crystal violet in the environment and aquatic products.The Ag NPs solution was prepared by the reaction of ammonium bicarbonate and silver nitrate,and the concentration of nano silver particles and R6 G were added to the slide chip,using multi channel experiment,low consumption can achieve rapid detection.By sliding the microfluidic chip to mix the two samples,then detected by SERS.The concentration of 10-10mol/L R6 G still have obvious Raman signal,the Raman shift is at 1360cm-1.With the passage of time,the intensity will change.The signal intensity changes in gradient,and exists the highest value.The signal intensity of 10-10mol/L was 400 cps.Continue to follow the crystal violet(CV)experiment,and successfully verify the high sensitivity of the surface enhanced microfluidic chip.(2)It presents a new method to fabricate 3D microfluidic chip using paper clip,microfluidic chip paper can adapt to a variety of different fluid channels.This method can be successfully applied to the rapid quantitative detection of bovine serum albumin and two valent iron ions.The linear range of bovine serum albumin is 5?mol/L-50?mol/L,the detection limit was 0.15?mol/L,R2 = 0.982.The linear range of two valent iron ion is 0.6?mol/L-12?mol/L,the detection limit was 0.18?mol/L,R2 = 0.992.Show clip 3D microfluidic chip can be used for paper based on possibility of actual sample testing.(3)Using SERS technology to complete the detection of tumor marker CEA on paper chip.First of all,SERS probe labeled CEA antibody was prepared.According to the specific binding of CEA antigen antibody,the "sandwich" immune structure was formed with the immobilized antibody and antigen attached to the chip.SERS was used to detect the MGITC signal of the antibody to be measured,and then the signal value corresponding to different concentrations of antigen was obtained indirectly.The concentration distribution of CEA in 1 ng/m L-20ng/m L,the concentration of molecular markers in the detection and the Raman peak intensity showed a good linear relationship,realized of high sensitive quantitative detection of tumor markers,CEA antigen detection limit is 0.3ng/ml.This method can be applied to the detection of more complex multi tumor marker protein,which has a bright future.
