In daily life due to excessive blood loss caused by casuaties accidents were common.It is the key to reduce the casualty rate.Rapid and effective hemostasis is the key to reduce casualty rate,which not only can relieve the pain of the wound,but also rescue the lives of patients timely.So the development of rapid hemostatic materials becoming the foucus of the study.Chitosan,Alginate and Bletilla striata polysaccharide as natural macromolecular polysaccharide have become the most popular research materials due to their good biocompatibility,biodegradability,absorbability,hemostatic property and wound healing.However the hemostatic property of single material was limited.In this article,in order to improve the hemostatic effect,the calcium alginate porous microspheres(Alg-Ca)with the function of molecular sieve were obtained by using emulsion crosslinking method and pore technique.Bletilla striata polysaccharide(Bsp)was extracted from the tubers by using water extraction and alcohol precipitation,and the composite hemostatic powder was prepared by combining with calcium alginate porous microspheres by freeze-drying technique.Finally,CS/Alg-Ca and CS/Alg-Ca/Bsp composite sponges were prepared by mechanical blending and lyophilization.And the structures and physical and chemical properties were characterized by SEM,particle size analysis,FT-IR,1H-NMR,biological microstructure assay,Zeta potential,water absorption,porosity and water vapor permeability,and the biocompatibility and hemostatic properties of the composite materials were evaluated by cytotoxicity,hemolysis rate,acute toxicity,RBC adsorption,PT assay,in vitro coagulation and animal bleeding models.The conclusions as followed:(1)The prepared Calcium alginate porous microspheres have large specific surface area and an open porous structure,according to the results of the water absorption,the best preparation condition was determined as follows: 2% alginate,5% CaCl2,0.5% porogen.(2)The results showed that the BCI value of the in vitro coagulation effect of Alg-Ca:Bsp=1:1 was the smallest and the hemostasis time was the shortest only 74.7s.The hemostatic effect was the best.(3)The physical and chemical properties,biocompatibility and hemostatic performance of CS/Alg-Ca and CS/Alg-Ca/Bsp composite sponges were characterized.The results indicated that the composite sponges with connected porous structure,and CS/Alg-Ca10 composite spongeswith strong water absorption,great porosity and higher water vapor transmission rate.In addition,the CS/Alg-Ca/Bsp ternary composite sponges were prepared on the basis of CS/Alg-Ca10 has good biocompatibility,in vitro coagulation and animal hemostasis test showed that CS/ALg-Ca/Bsp10 composite sponge had the shortest hemostasis time was 56.28 s and minimum blood loss was 0.024 g.So as a rapid hemostatic material is expected to be used in the filed of medical dressings.
