This paper makes research on the activity detection method of GOD and its effect on bread baking.First of all,I verified the process and data reproducibility of activity detection method which from different GOD manufacturers.According to the reproducibility,stability and detection time of the detection method,the o-anisidine coloration method in the continuous spectrophotometry method is improved.Based on large numbers of experiments,a rapid activity detection method was confirmed by data analysis of the reaction temperature was 37?,the reaction pH was 5.5,tha reaction time was 5 minutes,the dilution ratio of the sample was controlled to control the final dilution enzyme activity in 0.1u/mL to 0.5u/m L,substrate concentration was glucose solution of 0.3mol/L.Secondly,the optimal temperature was 25? to 60?,the optimum pH was 4 to 7,GOD has good tolerance under the condition of bread proofing temperature was 35 ?to 40 ?,pH was 5.0 to 6.0.Theoretically,GOD can meet the needs of bread baking process.At the same time,the effect of GOD on bread baking and the effect of secondary enzyme activity were analyzed by experimental system.The theory was verified when the addition amount of GOD is 0.2u/g,the addition of GOD can enhance the reticulate structure of dough,increase the flexibility of the dough and resistance to mechanical agitation,and increase the volume of the bread to make the sensory index of bread better.However,the dosage of glucose oxidase is not the higher the better,with the increase in the amount of glucose oxidase,its effect rise to a peak when show a downward trend.And higher content of bacterial alpha amylase was found through analysis of samples,a lot of residual sugar and dextrin was produced,so that the texture of the bread core is too soft and cannot support the expansion of the volume,resulting in bread volume too small.Therefore,it is necessary to confirm the activity of bacterial alpha amylase by detecting the enzyme before the practical application,and confirm the most cost-effective formula according to the practical application effect such as enzyme activity,bread formulation and baking process.
