The purpose of this study was to compare the composition,antioxidant activity and effects on diabetes of Lentinus edodes substrate polysaccharide(SLSP)and Lentinus edodes polysaccharide(LP).Firstly,the processes of extraction and deproteinization,and drying were optimized and the antioxidant activity of SLSP and LP was tested.The results showed that the optimum condition was hot water as solvent,solid to liquid 1:20,temperature 100°C,100 min and repeated 3 times,deproteinization by combination of sevag and enzymatic method,vacuum freeze-drying.Under these conditions,the yield of polysaccharides reached 13.00% and the molecular weight was 389.1 kDa.The composition of monosaccharide was arabinose: xylose: mannose: glucose: galactose in the tatio of 54: 26: 3: 11: 6.SLSP has better antioxidant properties than LP so it can be used as an antioxidant.Comparing the SLSP with LP and other spent mushroom substrate polysaccharide(SMSP),we found that the characteristics and composition of SMSP were related to fungi species and original component of mushroom substrate.This finding provide the basic data and theoretical reference for the production and application of SMSP.In order to explore the functional properties of SLSP and LP further,we tried to apply them to the treatment of Zucker diabetic fatty(ZDF)rats,but the effects of SLSP was not obvious.However,the therapeutic effects of LP on diabetes in ZDF rats with Impaired Glucose Tolerance stage administration(LI group)and diabetes mellitus stage administration(LD group)were significant,in which the effects of LI group was more obvious.Through the experimental study,the mechanism might be as following.LP regulated the distribution of gut microbiota and increased the relative abundance of butyrate producing bacteria such as Roseburia_faecis and Butyricicoccus_pullicaecorum to protect the L cell from inflammation.Therefore,the level of GLP-1 was improved and then the intestinal barrier was protected,so as to delay the occurrence and development of diabetes.
