Borage seed oil is a kind of small varieties rich in gamma linolenic acids special oil,it is rich in unsaturated fatty acid and fatty acid composition and contain a variety of physiological active substances,with the prevention and treatment of cardiovascular disease,lower cholesterol,and inhibit cancer and so on the many kinds of physiological health functions.At the same time,borage seed oil has been widely used in the fields of functional food and medicine because of its antioxidative effect.However,its anti-oxidative mechanism is still unclear.C.elegans(Caenorhabditis elegans)was known and highly conserved signal channel advantages and was widely used in life science field as a model organism,which has simple structure,short life cycle,gene sequence.C.elegans was used to explore the antioxidation mechanism for borage seed oil by optimized the soxhlet extraction process.And the main results for borage seed oil were as follows:1.Technological conditions of borage seed oil extract was optimum by single factor and response surface test.The result:extracting time 4.16 h,extraction solvent volume of 108.81 mL.extraction temperature 83.15?,under the condition,borage seed oil extraction rate was 34.05%,and the extraction of borage seed oil of the communist party of China identified 15 kinds of fatty acids,including 9 kinds of unsaturated fatty acid content was 81.27%,and the GLA content was 20.96%.The acid value of borage seed oil was 3.04,iodine value 150,saponification value 200,peroxide value 2.0,and the rotation rate of 1.4749.2.The life span of wild type of elegans was significantly prolonged by different concentrations of borage seed oil.the number of motor behaviors and subgenerations were increased.Among them,the fluorescence expression in the wild type of the 1%borage seed oil group was significantly decreased,indicating that the optimum antioxidant concentration of borage seed oil was 1%.Compared with the control group,wild-type caenorhabditis elegans after 1%borage seed oil treatment,motor frequency and spawning behavior were increased.nematode reduce the amount of stranded the active oxygen free radicals in the body,but its survival times,sports behavior and number of offspring was less than 0.1%GLA treatment group.3.1%borage seed oil processing daf-2 mutant worms swallow and discharge frequency increased significantly than wild type nematodes and the control group daf-2 mutant worms,heat stress significantly increased survival rate and survival time,and the body bent was un significant.The effect of 1%borage seed oil on daf-2 mutant nematode was more significant than that of wild nematode and control group daf-2 mutant.1%borage seed oil processing daf-16 mutant worms the survival rate was reduced and its life shorten than wild type worm,and 1%borage seed oil processing daf-16 mutant nematodes was daf-16 mutant worms body bending frequency increased significantly,stress the survival rate and survival time is reduced,but the oscillation frequency,swallowing and head does not affect the discharge frequency.At the same time,the expression of daf-2 and daf-16 in wild nematodes treated with 1%borage was significantly increased compared with the control group by using reverse transcription and real-time quantitative polymerase chain reaction.The effect of 1%borage seed oil on the antioxidant effect of daf-2 gene was demonstrated.4.The research of ethanol injection method,film dispersion method,ethanol injection,ultrasonic method and membrane dispersion,ultrasonic method,four methods preparation of borage seed oil injection of ethanol ultrasonic prepared liposome envelopment rate(99.99%)is significantly higher than the rest of the way,and the legal form of liposomes is relatively uniform.Using ethanol injection-ultrasonic method,borage seed oil in storage within 28 d stable liposome form,the envelopment rate had no significant change(p>0.05),storage rate over time was significantly increased(p<0.05).
