Due to the high sensitivity of surface-enhanced Raman scattering?SERS?and the high selectivity of immunochromatographic assay?ICA?,based on noble metal nanoparticles as substrates for immunoprobe and 4-mercaptobenzoic acid?MBA?as the Raman reporter,this thesis describes two kinds of high sensitive surface-enhanced Raman scattering immunochromatographic assay?SERS-ICA?for the determination of small molecular compounds.The main contents are as follows:1.Based on Au nanoparticles?Au NPs?,a SERS-ICA for multiple analysis was established.Two test lines?T line?were set on ICA paper for the simultaneous detection of two?-agonist?brombuterol and clenterol?and five fluoroquinolone antibiotics?enrofloxacin,norfloxacin,pefloxacin,ciprofloxacin,enoxacin?.Under the optimal conditions,the IC50 values of these compounds were 0.15,0.073,0.11,0.095,0.14,0.15,0.17 ng mL-1 and the corresponding limit of detection values were 0.41,0.94,0.30,0.39,0.46,0.41,0.51 pg mL-1,respectively.Swine meat samples spiked with different amounts of brombuterol and enrofloxacin were analyzed by the proposed method and the recoveries were found to be 88.7 and 108.3%with relative standard deviations?RSD?within 4.1 and 7.7%?for n=3?,respectively.2.Based on flower-like gold-silver core-shell bimetallic nanoparticles?AuNFs@Ag NPs?,an ultrasensitive SERS-ICA for the determination of brombuterol in swine meat and urine samples was developed.AuNFs@Ag NPs displayed strong SERS enhancement and the sensitivity of SERS-ICA based on Au NFs@Ag NPs was improved.Under the optimal conditions,the IC50 and LOD values were as low as 380pg m L-1 and0.5 pg m L-1,respectively.The cross-reactivity?CR?of the SERS-ICA with clenbuterol was only 8.5%and there was no CR of the SERS-ICA with other five compounds.Swine meat and urine samples spiked with different amounts of brombuterol were analyzed by the proposed method and the recoveries were found to be 95.8 and 108.0%with relative standard deviations?RSD?within 2.0 and 6.3%?n=3?,respectively.This thesis is focused on the studies of the SERS-ICA for multiple analysis and the preparation of novel noble metal nanoparticles?e.g.AuNFs@Ag NPs?with the high SERS enhancement effect for ultrasensitive detection of single analyte.The proposed two SERS-ICAs display the advantages from both SERS and ICA technologies such as simplicity,rapidity,low cost and ultrasensitivity,which make the SERS-ICA to be used for high sensitive detection of targe analyte on-site.
