At present,fluoroquinolone residues detection method requires strict laboratory environment,high hardware configuration and operation of high-quality personnel,long detection time,high cost,it's not suitable for screening a large number of samples,it is difficult to detect in the grass-roots units and promote the use of food enterprises.This study takes the fluoroquinolones mother nucleus structure of common characteristics of ciprofloxacin(CPFX)as hapten of monoclonal antibody technology,colloidal gold immunochromatography technology is introduced to the fluoroquinolones residue detection methods,and established a simple,rapid detection,high sensitivity,specificity and strong broad-spectrum fluoroquinolone drug residue immunoassay method,rapid detection of residues for clinical screening of fluoroquinolone antibiotics and a large number of samples,the animal food production process control,self-test provides a field test technology is simple and rapid,sensitive,convenient and reliable.The residues of fluoroquinolones in broiler samples from Hebei and surrounding areas were detected and analyzed.The status of Fluoroquinolones Residues in broiler samples from Hebei and surrounding areas was mastered,which provided technical support for monitoring and detecting Fluoroquinolones Residues.With the fluoroquinolones mother nucleus structure of common characteristics of ciprofloxacin(CPFX)and carrier protein(BSA)coupling,the preparation of artificial antigen immune CPFX-BSA,6 weeks old BALB/c female mice were obtained,the best sensitivity of broad-spectrum 4H8 monoclonal antibody,4H8 monoclonal antibody purified and labeled with colloidal gold particles as the gold standard pad.Using CPFX-BSA coupling protein and Rabbit anti mouse IgG as the test line and control,assemble the colloidal gold immunochromatographic residue detection of fluoroquinolone antibiotics.The test strip can be judged within 10 min by the naked eye.The 10 fluoroquinolones antibiotics were all positive,while 4 other common drugs were negative,indicating that the paper was a good test specificity for 10 fluoroquinolones antibiotics.The detection of ciprofloxacin,pefloxacin,danofloxacin,ofloxacin,enrofloxacin,norfloxacin,lomefloxacin limit was 3.2?g/kg,detection of salad difloxacin,enrofloxacin,marbofloxacin limit was 6.4?g/kg.The stability test shows that the paper strip can be kept stable at 60 d at 37?,and can be kept 360 d at 4?.Using the test strip detection of the 3414 chicken samples in Hebei province and the surrounding areas,the average positive rate was 0.76%,including slaughterhouses,supermarkets,live poultry market average positive rate were 0.38%,0.09%,2.19%,and ELIAS is on the same test results.It shows that the colloidal gold immunochromatography paper developed in this study can be used for rapid screening and detection of fluoroquinolone residues,which is convenient to be popularized and applied at the base.
