Molecular imprinting technique(MIT)refers to a technique in which template and functional monomer forms a pore polymer by covalent or non-covalent binding under certain conditions.The molecularly imprinted polymer has a hole which is consistent with the template in spatial structure,thus has an ability of specific recognition of the template.Molecular imprinting techniques have been widely used in many fields such as solid phase extraction,membrane separation,chromatographic separation,biosensors and drug analysis.The surface molecularly imprinting technique is a technique that molecularly imprinting occurs on the surface of substrate,then the spatial hole is out of the polymer,the recognition sites is exposed at the surface of substrate.Molecularly imprinted solid-phase extraction(MISPE)is a method using molecularly imprinted polymers as the sorbents of solid-phase extraction to enrich and purify target molecular.MISPE is a more efficient way for the separation and enrichment of target molecules from complex samples based on the highly selective adsorption of molecularly imprinted polymers.MISPE plays a significant role in the separation of active ingredients from the traditional Chinese medicine,natural products and biological samples.Lycorine and Xanthopterin were selected as the research objects because they are closely related to cancer.Lycorine can inhibit the tumor cells and have a certain value in clinical research.Xanthopterin,as a potential marker of cancer plays an important role in the cancer diagnosis.However,since the content of lycorine in Crinum asaticum L.var.sinicum and Xanthopterin in the urine is relatively low,using the general separation method have many difficulties.In order to solve the problem,a new method of molecular imprinting technology combined with solid phase extraction was put forward for separation and purification.The mainly research aspects are as follows:1.Using silica nanoparticles as the matrix of preparing surface molecularly imprinted polymer,core-shell type lycorine molecularly imprinted nanospheres were synthesised.The nanospheres were charactered by transmission electron microscope scanning and IR.The result shows that the success of preparation of lycorine molecularly imprinted polymers.The adsorption equilibrium and adsorption kinetics were researched,the molecularly imprinted polymer by Scatchard analysis in the process of adsorption shows that there are two types of adsorption site,the imprinting factor is 2.5.The result shows that the molecular imprinting polymer has the ability to recognize the target molecule with special recognition.A new method is provided for the purification of trace elements in the complex components of Chinese medicine.2.The silica nanoparticles were modified with vinyl,and the infrared spectra indicate the success.The nanospheres were synthesized using xanthopterin as the template at the surface of silica nanoparticles.The nanospheres were charactered by transmission electron microscope scanning,which showed the nanospheres were successfully synthesised.The adsorption performance results showed that the imprinting factor is 7.8,which indicates that the surface molecular imprinting nanospheres have specific adsorption of xanthopterin.The nanospheres were used as the sorbents of solid-phase extraction,high performance liquid chromatographic fluorescence detection method was used to detect the xanthopterin in the urine of the tumor rats.It provides a new method for detecting target molecules in biological samples.
