Bioimprinting is one of the most promising new methods for overcoming cancer targeting defects.Using polymer materials to create highly fidelity imprinted substrates for template identification,combining structurally complementary functionalized polymer monomers with non-covalent or covalent bonds to the template,followed by the addition of a cross-linking agent for polymerization After the reaction is completed,the template is eluted to form an imprinted substrate having a fixed concave size and shape and a specific alignment functional group.The imprinted substrate and the template molecule have excellent complementarity in terms of shape,size,and the like.The polymer has the function of specifically recognizing the original template.The early related research mainly focused on the molecular level,but its use for molecular recognition of bioactive components of the components was prone to deactivation and was limited by the complexity of the template size,so no cellular dimension was involved.Based on this,a highly fidelity cell-imprinted polymer was fabricated in this study.Cells with complex structures can be captured,and the properties of imprinted polymer membranes can be further explored at different levels.At the same time,a detection device for rapid capture and separation of target cells was created.Expanded the application of imprinted polymer membranes in the field of biological detection and sensing.1.Construction of a novel cell imprinting model and study of its properties.Polydimethylsiloxane has excellent physiological inertness,resistance to heat and cold,non-toxic anti-seismic and other properties,and has no side effects on cell tissues,in line with the requirements of biomedical materials.In this work,we use polydimethylsiloxane The bulk of the alkane and the curing agent produce a highly fidelity cell imprinting polymer.The cell imprint produces an artificial receptor that captures the target cell by specific size and shape of the recessed structure.Characterization under an atomic force microscope shows a highly symmetrical structure with the template cells.We report here that the cells are inactivated by a combination of dialdehyde fixations.The advantages are:(1)hardening of the cell surface by cross-linking amine groups with aldehyde groups;(2)eliminating chemicals secreted by living cells and preventing interference with cellular imprinting The fidelity of the formation process.The target cell capture efficiency is 89%,and highly anti-interference.At the same time,this study provides a simple and inexpensive single-cell isolation model,which does not rely on professional or expensive equipment,does not require affinity reagents or protein antibodies,and provides a wide range of single cell analysis,cell differentiation and cell-based clinical diagnosis.2.A new method of tumor cell recognition and detection based on cell imprinting.In this work,on the basis of the fabricated cell bioprint polymer film,further studies were performed on its apparent single molecule properties and as a sensor mechanism.The high-fidelity imprinted polymer membranes produced were connected to the Ligprobe via the folate receptor and then characterized by RCA rolling-loop amplified signal amplification,demonstrating the high degree of agreement between the imprinted polymer and the template cells at the micromolecular level;Distinguishing the overall size and surface characteristics of target analytes on the nanometer scale,due to the difference in nanoscale,biobleaches have the ability to distinguish cells on the micrometer scale,which means that the imprinted polymer film may be used as a biosensor.Can be used to separate cells based on cell surface membrane protein species differences,this difference may be used as a biomarker-based cancer diagnostic tool,this study also made based on imprinted polymer membrane electrochemical sensing device,the device is low-cost,easy to operate Can be used repeatedly,has a very broad application prospects in the field of biological sensing.
