Selenium,a trace element essential to human beings,has good anti-oxidation and antitumor functions.In recent years,selenium has been rapidly developed in the field of life science due to its excellent biological activity.Bamboo leaf extract has good anti-oxidant,antihypertensive,antitumor,immunomodulatory and other activities,and it has also been extensively studied due to its cheap and high yield;grease has been one of the six essential nutrients and is one of the three major energy-producing nutrients.In this paper,we use sodium selenite,selenocystine,bamboo leaf extracts and their compounds as the study objects,extract the bamboo leaf extraction by extraction method,optimize the levels of extraction factors by orthogonal method and scan and analyze the functional groups of bamboo leaf extraction in the wavelength range of 4000 to 400 cm~-11 by potassium bromide pressed-disk technique.Then,we use centrifugal sedimentation rate and effect of inhibiting peroxide value as indicators to screen emulsifier and adopt oven drying method to study the oxidation stability evaluation on three kinds of grease(rapeseed grease,lard and idesia grease).On this basis,we adopt CCK8 method to study the inhibition of Hela cell proliferation in vitro with the Hela cell as a cell model.We hope that it can inhibit grease oxidation as a composite antioxidant,at the same time supplement selenium as food and play a rule in anti-cancer and adjuvant treatment of cancer.The main study results in this paper are as follows:1.We extracted bamboo leaf extract of moso bamboo by the extraction method,conducted single-factor test with extraction temperature,extraction time,solid-liquid ratio and ethanol concentration as the factor variables.Then we adopted spectrophotometry to determine the yield of bamboo leaf extract and studied the effects of different levels and different variables on the yield of bamboo leaf extract.On the basis of the single-factor test,we conducted the orthogonal test.At the same time,we used SPSS20 statistical software to conduct range analysis on the test results so as to determine the best test combination.From the test results we can see that when extracting bamboo powder for 150min at 90?by the solid-liquid ratio of 1:40in 60%ethanol,the highest yield was 1.71%.2.We adopted sodium thiosulfate titration method to determine the peroxidation value and used peroxide value as an indicator to evaluate the oxidation degree of grease.At the same time,we adopted oven drying method to study the oxidation stability effects of sodium selenite,selenocystine,bamboo leaf extracts and their compounds on rapeseed grease,lard and idesia grease.The results showed that sodium selenite,selenocystine,bamboo leaf extracts and their compounds had certain antioxidant effects on rapeseed grease,lard and idesia grease.The antioxidant effects of the three kinds of grease kept consistent and existed positive-correlation dose-effect relationship.At the same time,the compound antioxidants showed the synergistic anti-oxidant effect.The ability of anti-oxidation of grease from weak to strong was as follows:sodium selenite<selenocystine<bamboo leaf extract<bamboo leaf extract+sodium selenite<bamboo leaf extract+selenocystine.The effect of compound antioxidants of bamboo leaf extract and selenocystine(0.05 g+500?g)was best,close to TBHQ(0.02%)anti-oxidant effect,which proved its potential to be developed as the natural complex antioxidant.3.We adopted CCK8 method to study the inhibition effect of sodium selenite,selenocystine,bamboo leaf extracts and their compounds on Hela cell proliferation in vitro.The results showed that all of the 5 antioxidants showed certain anti-tumor activity.The growth inhibition ratio of the other 4 antioxidants except bamboo leaf extract to Hela cells had positive correlation with dose.400?g/mL of sodium selenite,400?g/mL of selenocystine,100?g/mL of bamboo leaf extract,400?g/mL of sodium selenite+1000?g/mL of bamboo leaf extract,400?g/mL of selenocystine+1000?g/mL of bamboo leaf extract could reach their maximum inhibition ratios at 48 h,and the maximum inhibition ratios were as follows:86.53%,79.02%,11.16%,89.54%and 82.18%respectively.
