Lutein is a kind of natural pigment which can be used as feed and food additives,which can also prevent some diseases and have many effects of pigment and the pigment is not in the human or animal body what can only be get from nature synthesis.Chlorella has many advantages such as fast growth and cultivation conditions of easy control and high biomass.The heterotrophic chlorella is Tianguan Group introduced to get high oil.The heterotrophic chlorella before is mainly used for the production of biodiesel,and the lutein as a high value-added products,the extraction is very necessary,which not only protect the environment but also help resources charge Separate utilizationIn this paper,we study on the process of the micrococcosis and the extraction of crude lutein.1.This experiment by heterotrophic chlorella as raw material,using enzymatic wall-breaking extract rough lutein,by optimizing the process conditions to improve coarse lutein yield and lower production cost,which provide the basis for industrialized production.By single factor experiment,the coarse lutein yield influential factors digestion time,liquid-solid ratio and adding amount of three aspects has carried on the investigation,and on this basis,through the Central Composite Design(CCD)response surface method to Design the three factors three levels to optimize the experiment.According to the analysis of design-expert 8.0.5,the optimal experimental conditions were 2 h for enzyme solution time,6.31 for liquid solid and6.21 for enzyme,and 20.18% for the best crude lutein production.According to the regression equation and the actual experimental operation,the enzyme solution time was selected,and the liquid solid ratio was 6,the enzyme was added 6 mL,and the yield of crude lutein was 20%.2.In extraction process,the experiment using single factor experiment on theinfluencing factors of extraction of crude lutein extraction time,extraction temperature,ethanol and ultrasonic shock power four aspects has carried on the investigation,on this basis,according to Box Benhnken Design(BBD)Design principle of extraction time,extraction temperature,ethanol content on the three factors three levels of response surface optimization Design of experiment.The optimal experimental scheme was obtained by the Design-expert 8.0.5 software analysis and the optimum experimental conditions were designed based on this.The optimal experiment scheme for extracting temperature 50.95℃,ethanol content 61.47 mL,extraction time 1.03 h,coarse lutein yield forecast of 21.67%.According to the design of the optimum experiment conditions for the extraction temperature 51℃,amount of ethanol to add 61 mL,extraction time of 1 h,the experiment measured the coarse lutein yield is 21%.In this experiment,compared with the previous experimental,there are some innovations: first,the material is different,in this experiment the heterotrophic chlorella is from Tianguan Group which has high oil content of heterotrophic chlorella,which initially is only used for biodiesel extraction,and in this experiment is used as raw material to extract crude lutein,in this way we may improve the added value of the algae,thereby reducing the cost of production.Second,through the optimize of the test method,combined with the high oil content of heterotrophic characteristics,the experiment is carried out through single factor test,final test plan and the test conditions and optimized by response surface method.
