Bifidobacterium is a beneficial probiotic for human health.Bifidobacterium has a variety of physiological functions.It is able to regulate the body immunity,lower cholesterol,improve antioxidant and so on.Due to the limitation of anaerobic culture conditions,it makes it difficult to play a physiological function.Therefore,the researches on improving the number of viable Bifidobacteria are more and more.It is more important that it can improve the number of viable Bifidobacteria.In this experiment,the tomato juice,carrot juice and corn peptides were added to optimize the composition of the medium.The research mainly explored optimization of fermentation conditions for Bifidobacterium by response surface methodology,based on Design-Expert software design.The contact of Bifidobacterium,corn juice concentration,pH and fermentation temperature were investigated on viable count.The methods for immunity function were relative to weights of spleen and thymus,carbon expurgatory index,plaque forming assay,DHT,and Elisa.The effect of mixed probiotic cultures on the intestinal microbiota of male pandas was analyzed by high-throughput sequencing in this investigation.(1)Based on the single-factor test,growth-promoting factors were optimized.The best growth-promoting factors of Bifidobceterium Longum-01 were 11.6% of the tomato juice concentration corn peptide,9.4% of the carrot juice concentration and 1.7% of the corn peptide.The best growth-promoting factors of Bifidobceterium adolescent-10 were 13.0% of the tomato juice concentration,11.0% of the carrot juice concentration and 1.0% of the corn peptide.The best growth-promoting factors of Bifidobceterium animalis-30 were:11.0% of the tomato juice concentration corn peptide,10.7% of the carrot juice concentration and 2.0% of the corn peptide.(2)Based on the BBD test principle,the culture conditions were optimized.The optimum culture of Bifidobceterium Longum-01 were obtained: 4.9% of the Bifidobceterium Longum-01,12.1°Bx of the corn juice concentration,pH7.0,and fermentation temperature of 39.2?.The optimum culture conditions of Bifidobceterium adolescent-10 were obtained: 5.0% of the Bifidobceterium adolescent-10,14.0°Bx of the corn juice concentration,pH 6.5,and fermentation temperature of 38.8?.The optimum culture of Bifidobceterium animalis-30 were obtained: 7.4% of the Bifidobceterium animalis-30,12.0°Bx of the corn juice concentration,pH 6.5,and fermentation temperature of 37.4?.(3)Through the carbon expurgatory index,complex probiotics and complex probioticscorn peptide have obvious changes compared with the control team.Through the splenic lymphocyte proliferation experiment,complex probiotics and complex probiotics-corn peptide have markedly obvious changes compared with the control team.The experiment showed that complex probiotics and complex probiotics-corn peptide enhanced certain parameter of splenic lymphocyte function.Through DHT experiment,the results implied that complex probioticscorn peptide have changes compared with the control team.Through the Elisa experiment,the experiment implied that complex probiotics and complex probiotics-corn peptide could enhance IgA.In conclusion,complex probiotics and complex probiotics-corn peptide could be able to enhance mice' immunity function.(4)Through high-throughput sequencing,the results could show that complex probiotics can improve Simpson index and lower Shannon index.Complex probiotics could be able to motivate intestinal microbiota imbalance.
