| Gelatin,which comes from animal bone,tendon,cartilage,epimysium and hide,has been widely utilized in food,medicine,cosmetic and other industries.Animal bones,as a by-product of processing,have not been fully used.This paper studies the process of extracting gelatin from animal bones and its physico-chemical characters,which is not only reduces environment pollution but also is of great significance and economic value.First,the minerals in animal bones are removed by acid.The hydrochloric acid concentration,ultrasonic power,pickling time,pickled material liquid than minerals for the influence of removing minerals are studied.The bone is desalted with HCl on the best conditions as follows:HCl concentration=3%,ultrasonic power=1000W,bone :HCl=30:300(g/ml),pickling time=72h.Then,the influence of the alkal leaching time,ultrasonic power,and water extraction pH,water extraction material liquid ratio is studied under the condition of the best removal of minerals.The best condition for the gelatin extraction yield is certained as follows:alkali leaching time=6d,ultrasonic power=1000W,pH=5,bone:water=3:10.The gelatin extraction yield is to 14.52% with the best conditon.The analysis of physical and chemical properties of gelatin mainly is studied.The distribution of gelatin molecules ia mesured by SDS-PAGE,while the structure of gelatin is analyzed by ultraviolet absorption spectrum.The SDS-PAGE electrophoresis figure shows that quality of gelatin molecules is dispersed,without a certain value but more than20.0kDa.The ratio of extraction of the Protein is different at different temperature.The ratio of extraction of the protein is higher at the temperature of 60℃,70℃,80℃.However,the ratio of extraction of the protein is relatively less at the temperature of 50 ℃ and90 ℃.There is a strong absorption peak at 220 nm and a weak bench at 280 nm in the ultraviolet absorption spectrum of gelatin,which is explained aromatic amino acid content in gelatin is less.This research provides a theoretical basis for extracting gelatin from the animal bones.It not only offers a new method for the comprehensive use of animal bones,but also provides a source of raw materials for gelatin production industry. |
