Crude oil-contaminated soil samples were collected from Dagang Oilfield.The medium was enriched and domesticated to cultivate crude oil degrading bacterial flora.Flat scribing could separate crude oil degradation bacterial flora.Crude oil degraded mixed bacterial flora could be constructed using single strain of crude oil degraded.The mixed bacterial flora were immobilized.The degradation mechanism of crude oil studied could analyze the degradation characteristics of the free flora and immobilized flora to the components of crude oil.Related research findings are as follows:1.Artificial seawater medium containing crude oil was used.Three stages of domestication and cultivation were carried out.The crude oil degrading bacterial flora were isolated from the soil of Dagang Oilfield.2.Six strains of crude oil degradation bacteria were screened out after domestication and cultivation.The six strains of crude oil degradation bacteria were respectively Lysinibacillus,Bacillus flexus,Pseudomonas mendocina,Pseudomonas alcaligenes,Bacillus thuringiensis and Pseudomonas alcaligenes.The degradation rates of crude oil were lower for the six single strains.3.Six strains of single bacteria screened out could construct as mixed bacterial flora.The mixed bacterial flora was named as YJ01.And the degradation rate of bacterial flora was determined.YJ01 mixed bacterial flora had a better degradation effect on crude oil after 14 days of degradation.The degradation rate of YJ01 mixed bacterial flora was 80.64%.4.Gas chromatography could be used to study the YJ01 mixed bacterial flora on the degradation of crude oil alkane mechanism.The degradation rate of medium alkanes was the highest after 14 days of degradation.The degradation rate of medium alkanes was 81.88%.YJ01 mixed bacterial flora had different degradation characteristics for different kinds of alkanes in saturated hydrocarbons.YJ01 mixed bacterial flora had good degradation effect on odd carbon alkanes,high carbon alkanes and phytane in branched hydrocarbons.5.Mixed bacterial flora immobilized by ramie had good degradation effect on the crude oil.The 7-day degradation rate of mixed bacterial flora immobilized by ramie was up to85.19%.The degradation effect was stronger than the activated carbon immobilized bacterial flora and the non-immobilized free bacterial flora.The carbon number of the main peak migrated obviously after immobilized by ramie.Bacterial flora immobilized by ramie to even carbon alkanes and lower carbon alkanes had stronger degradation advantage.Bacterial flora immobilized by ramie showed better degradation of pristane.It could provide a theoretical basis for the degradation of high pristine crude oil.
