Manganese peroxidase?MnP?is a glycosylated extracellular peroxidase,which has the special ability to degrade lignin and aromatic compounds,and has a significant effect on the decolorization of the dye.In order to improve the activity of MnP in the strain TN02A7-Lg-mnp2,the culture conditions of TN02A7-Lg-mnp2 and MnP enzyme were optimized and the conditions were optimized.To the actual production,in the optimized culture conditions of the transformant strains of the dye decolorization of the study.The results of optimization and induction experiments show that the pH value of sodium malonate buffer solution has a significant effect on MnP activity in MnP enzyme detection system.At pH 3.5,the highest amount of MnP was observed.The optimal culture conditions of TN02A7-Lg-mnp2 were as follows:the optimum culture conditions were as follows:the optimum rotation condition was 220 r The concentration of Mn-1 and Mn2+was 267?mol·L-1,the pH value was 6.5,the concentration of heme was 0.05g·L-1,the optimized enzyme activity was about 91U/L after optimization,The enzyme activity was improved by about 9 times.And then the six inducers that affect the enzymatic activity of MnP were screened to obtain the inducer that could increase the activity of Aspergillus nidulans transformant strain TN02A7-Lg-mnp2 MnP as wood chips,followed by gallic acid,benzoic acid,resveratrol and wheat bran,The worst effect of vanillin not only did not improve the activity of MnP enzyme,but inhibit the enzyme activity.The results of decolorization of the dye showed that the enzyme solution of the transformant strain had decolorization effect on the six kinds of dyestuffs,malachite green,neutral red,Congo red,lute blue and crystal violet.The decolorization rate of the optimized enzyme was 98.36%,and the decolorization rate was 98.36%,respectively.The decolorization rate of the enzyme was 98.36%,and the decolorization rate was 98.36%and 96.47%respectively.The decolorization rates of Congo red and Quilben blue were 96.76%and 89.39%,respectively.The decolorization rate of crystal violet was 67.31%.The results showed that the decolorization rate of turquoise blue and Congo red was 74.77%and 63.24%,and the crystal violet reached 57.83%when the concentration of malonite and neutral red was 100%,But Dan Dan in 16h only reached 38.16%.The results showed that the extracellular enzyme of TN02A7-Lg-mnp2 had a significant decolorization effect on malachite green and neutral red by two different methods.The crystallization of purple and Dan decolorization degradation effect in general.
