In order to investigate the effects of different concentrations of linseed oil and condensed tannin on the duodenal's structure and PepT1 expression of Hu lamb,30 weaned Hu lambs were selected and fed.After being slaughtered,hematoxylin-eosin(HE)staining was used to observe the morphological changes of the duodenum.Scanning electron microscopy was used to observe the morphological changes of duodenal microvillis.Immunohistochemistry and western blotting were used to detect the distribution and expression of PepT1,which was analyzed by bioinformatics.Results:1.The thickness of duodenum in the case of adding only condensed tannin without adding linseed oil,the thickness of the duodenum of Hu lamb increased with the addition of the concentration of condensed tannin.When 4% linseed oil and different concentrations of condensed tannins were added simultaneously in the diets,the thickness of the duodenum of the Hu lamb also increased.2.The thickness of the mucosal layer of the duodenum of Hu lamb increased with the addition of the amount of condensed tannin without adding 4% linseed oil.When 4% linseed oil was added,duodenum mucosal thickness of Hu lamb had increasing tendency with the addition of condensed tannin concentration to increase.3.The length of microvillis in duodenum of Hu lamb was significantly different from the addition condensed tannin's concentration of 0.2% and 0.4%.There was no significant difference between the addition of condensed tannins 0% and 0.4%,respectively.After adding 4% linseed oil,there was no significant difference in the length of duodenal microvillus with the increase of the concentration of condensed tannin added in the feed,but there was a decreasing trend.4.The Pep T1 mainly expressed in duodenal absorptive cells in Hu lamb.Without the addition of linseed oil,the expression of Pep T1 increased with the increase of condensed tannin concentration.Compared with the addition of 4% linseed oil and condensed tannin in other group,the expression level of Pep T1 was the highest after adding 4% linseed oil and 0.2% condensed tannin in the feed.The possible reason was that the increased expression of fatty acids in the duodenum after the addition of linseed oil affected the expression of Pep T1.Similarly,western blotting showed the same results.5.The Pep T1 sequences of 5 herbivores and Hu lamb were selected for homology analysis,it was found that the homology of PepT1 between Hu lamb and Ovis aries was 99.9%,the homology of PepT1 between Hu lamb and Capra hircus was 98.8%,the homology of Hu lamb and Bos Taurus and Bos grunniens was all 96.2%,and the lowest homology with Oryctolagus cuniculus was 80%.From the sequencing results,the nucleotides of Hu lamb and Ovis aries had two different places,which was 1,673 th and 2,013 th respectively.At 1,673 th,Hu lamb's was G,and Ovis aries was C.Then the nucleic acid sequence was translated into the amino acid sequence and showed that the amino acid at position 558 th changed,which the Hu lamb's was S,and the Ovis aries was T.Although there was a change in nucleotide at 2,103 th,there were no changes in translation into amino acidsConclusions:1.Adding 0.4% of condensed tannin to the feed was the most suitable for small peptide transport.2.The feed of addition of 4% linseed oil and 0.2% condensed tannin was most suitable for the growth of Hu lamb.3.Compared with the Ovis aries,the 558 th amino acid of PepT1 in the Hu lamb duodenal enterocyte was changed and the amino acid was located between the extracellular domain.
