Aflatoxin B1(AFB1)is a highly toxic mycotoxin,widely found in various agricultural products,and has a strong carcinogenicity and is listed as a group I carcinogen.Currently,the methods for detecting AFB1 mostly need professional operation and expensive requirements,is unable for rapid and qualitative detection for AFB1.The development of high accuracy,high sensitivity and onsite methods of detecting AFB1 is of great significance to food safety.Among the all disc storage technologies,Blu-ray Disc(BD)Technology has the largest storage capacity and the smallest focus diameter.Beyond the intrinsic capability of storing and archiving high-definition audio-video and high capacity data software,it has been adopted to analytical chemistry and molecular detection field.BD/BD drivers' system has the potential to be the simple and portable on-site detection methods with high sensitivity,high accuracy and low cost.In this paper,we mainly researched the quantitative detection of AFB1 based on BD technology.BDs was activated and adapted as substrates to prepare an indirect competitive immunoassay with a microfluidic channel plate.The error data of the assays on BDs is read by an unmodified Blu-Ray drive combined with a free disc-quality check program(Plex Utilities)for the quantitative detection of AFB1.The main research contents are as follows:1.The activated BDs was adapted as substrates to prepare immunoassays with a microfluidic channel plate.The signals of the assays were enhanced by silver staining and the binding strips presented on BD are corresponding to the AFB1 concentration.2.We have developed a biomolecular detection platform based on BD technology.BD adopts error correcting codes and modulation codes to verify the errors.A standard drive controls laser head tracking and reading data.we realized the diagnosis for the microarray on BD using the error correcting mechanism of BD and the precise servo control principle of the laser head.3.The BD detection system have achieved the qualitative detection of AFB1 in corn samples.The detection range is 0.5-200 ng/mL,the limit of detection is 0.5 ng/mL.Compared to commercial ELISA kits and traditional HPLC,the test results confirm the accuracy and reliability of this method.
