Spectral domain endoscopic optical coherence tomography imaging technology is a new biology endoscopic technology in bio-medical field. This technology has advantages such as high spatial resolution, high imaging speed and so on. It can rapidly imaging for annular or cavity tissues and organs and widely used in today's medical surgery. Endoscopic OCT probe design and fabrication has some problems such as imaging depth and lateral resolution restraint with each other. In the endoscopic OCT probe design based GRIN lens,we make a detailed mathematical analysis for the optical path model and use Gaussian optical ABCD transmission matrix to the establish the input and output functions of the probe spot diameter and Gaussian complex parameter q. We obtain the working parameters of the relationship probe and the conclusion that the gap length between GRIN lens and single-mode fiber affect obtained probe will affect the output of the spot diameter and working distance; and accordingly developed a probe design process; use the linear CCD probe to measure the actual working distance of the output light, the experimental results show good collimation properties of the probe, the lateral resolution of the system is high.Papers have studied the spectral domain OCT system data acquisition and control technology in detail, analyzes the key parameters of spectral analysis, the scope of work, the spectral resolution, etc., based on the signal affected by the parameters above, we design and construct the spectroscopic system; the influence of image acquisition module frame grabbers and acquisition speed linear CCD, the design of technical route synchronization control, use signal generator or synchronization signal acquisition card and application DAQ synchronous acquisition outputs a control signal; Labview-based control acquisition software systems is set up, experiments verify the performance of the system.
