Autologous fat tissue,with the advantages of a rich source,good filling shape,small postoperative trauma,perfect tissue compatibility and simple operation,has been considered as the main source of cells in tissue engineering,regenerative medicine and cell therapy,and which is applied widely in other fields.However,there are also the problems of low survival rate of adipose cells and postoperative complications for autologous fat grafting.Purification of the lipoaspirate,an important step in the technique of adipose grafting,has a far-reaching effect on improving survival rate after transplantation.The common purifying methods,such as centrifugation and precipitation,have some advantages and shortcomings in terms of efficiency,cost,impurity separating,activity and density of purified fat particles etc.,which has no uniform standard in clinical practice.Meanwhile the risk of fat cell contamination is increased due to the open processing environment.Therefore,this paper presents a process flow for purification and homogenization of autologous fat and develops the purification and homogenization instrument of autologous fat,to achieve autologous fat’s closed automatic purifying and homogenizing treatment,and promote the standardization study on purification of fat.The details are as follows:⑴Based on the review of the research status of autologous fat,adipose stem cells and adipokines,as well as the analysis of the survival rate of fat granule grafting and the processing method of fat in vitro,the overall program and structure of autologous fat purification and homogenization instrument have been proposed and developed,of which the fat handling container unit was designed specifically.Furthermore,the purification and homogenization process flow of autologous fat was put forward.⑵This paper discusses functional requirements of the instrument for autologous fat’s purification and homogenization,and clarifies hardware platform system solutions,and corresponding hardware control system was realized in a way of modular development,including ARM microcontroller module,motor driving module,signal feedback module,man-machine interaction module and power module.And in the process of development,the relationships among modules,the reasonable choices of electronic devices,parameters analysis as well as circuits design and implementation were fully considered.⑶Accomplish study and transplantation of μC/OS-Ⅱ operating system and μC/GUI graphics library.Then based on the STM32F10 x library function programming,the whole framework of autologous fat purification and homogenization automatic control software system has been designed rationally,in addition,the overall scheme of corresponding software control system has been defined,and the major module development was completed,including the application,STM32 underlying configuration,user interface and interrupt handling,etc.And the entire software control system fully adapts to the corresponding hardware system.⑷The hardware debugging,machine assembling and software debugging of autologous fat purification and homogenization instrument were accomplished,and the purification and homogenization instrument is stable and reliable as carrying out simple preliminary experiment,which can meet the functional requirement of adipose grafting purification.In summary,the device,with reasonable structure,short development cycle,high scalability,low maintenance costs,high reliability in hardware and simple interface,strong real-time property and maneuverability in software,is designed and developed to provide a new method and apparatus for autologous fat transplantation,and which is expected to lay a foundation for the standardization and clinical application of fat purification.
