| Large yellow croaker(Pseudosciaena crocea)is a drum fish(Sciaenidae).Zhoushan Island is one of the main productive locality in Zhejiang Province for large yellow croaker.Recently,the rapid economic growth in the coastal areas as well as the expansion of human population leads to the depletion of marine fishery resources and deterioration of environment.Overfishing has caused a significant reduction towards the wild large yellow croaker resources and eventually leads to the loss of fishing season.The market supply of large yellow croaker mainly depends on the stocks cultured in the hatchery or in a aquaculture system in either intensive or extensive.Culturing large yellow croaker include problems that are commonly known as germplasm degradation,precocious puberty,and high disease susceptibility.In this study,we use SLAF sequencing technology to screen thousands of SNP markers from the large yellow croaker genome.Through the analysis of genetic variation,30 individuals were selected for the maximum body size and 30 additional individuals were selected for minimum body size.These individuals are all derived from the same generation.Moreover,the analysis of linkage is approached by applying the quantitative trait loci(QTL)tags that are correlated to the size and the growth of large yellow croaker.Thus,the QTL has been developed.The original number of SNP is 153,406.However,there are 151,253 SLAFs after being filtered.The average depth is 6.68 x.According to the SNP SLAF detection,SNP loci are screened and the SLAF polymorphism is 65.88%. |
