Effect Of IL-34 In The Progress Of JEV Infection

Epidemic encephalitis B is a zoonotic infectious disease caused by Japanese encephalitis virus(JEV). Selective infection of the central nervous system occurs when the patients infected with JEV for about four days. Compared with the other infectious diseases, epidemic encephalitis B has serious consequences with high mortality, and severe sequela, but there is no effective treatment in clinic, still focuses on prevention. Recent studies have found that IL-34, secreted from nerve cells in the central nervous system, can activate microglia through specific CSF-1R(Colony-stimulating factor-1), and further reduce the nerve damage caused by oA?(amyloid beta protein), the study reveals that IL-34 plays a neuroprotective role through regulation of microglia. To date, the role of IL-34 in JEV infections remains unknown. Thus, to determine the role of IL-34 in JEV infections, first we conducted in vivo experiments on C57BL/6 mice, 3-4 weeks old mice were inoculated JEV(104 CFU). Surprisingly, mortality of IL-34 group mice is much higher than the PBS group. the mouse brain grinding after inspection of the virus titers were found, and IL-34 group of rat brain virus titer also far higher than the PBS group, three repeated experiments has been the same. In addition, in vitro experiments based on microglia cells as model cells, adding group virus titer of IL-34 is higher than that of the supernatant of PBS cells group. In a summary, the study found IL-34 in JEV infection process can promote the replication of the virus, but IL-34 promote JEV infection mechanism needs to further research and nerve disorders that occur in the process of new features, to shed light on the interaction between neurons and microglia, widening to JEV infection and pathogenesis of cognitive, on the development of new and effective therapeutic method provides a new way of thinking.
