Based on maize straw returning for successive years of soil samples as bacteria source,screening strains capable of degrading cellulose,combined with high protein,high lipid exogenous bacteria,degradation by strain between antagonism experiment building composite strains ZW-1,through the liquid and solid fermentation experiment strains of CMC enzyme activity,filter paper enzyme live live live,pro-tease and amylase of rotten composite strains ZW-1to evaluate the degradation of function;The other strains through the analysis of the microbial diversity detection method of ZW-1stability and microbial community structure;Finally through the laboratory simulation of field experiment and field trials to evaluate the application effect of compound strains ZW-1.The main results were shown as follows:1)Isolated from soil of perennial straw counters-field screening of cellulose degradation strains,the CMC-Congo red staining method measuring cellulose hydrolysis circle diameter on the separation filter,and by measuring the strain of cellulose enzymeactivity and filter paper enzyme activity after screen,cellulose decomposing bacteria for selected from 5 strains(XJ1,XJ2,XJ3,XJ5,ZJ2)for subsequent research.2)According to the liquid fermentation strains pro-tease and amylase activity in size,from the laboratory existing strains in the screening of enzyme production ability of high strain: Bacillus subtilis,Bacillus licheniformis,Paenibacillus mucilaginosus,Bacillus amyloliquefaciens,Bacillus cereus,Aspergillus oryzae,Aspergillus niger,Streptomyces microflavus,Lactobacillus acidophilus,Saccharomyces cerevisiae.3)Bacteria bread and Oxford cup method is adopted to improve the experiment of antagonism,decomposing and maize straw and will get 5 strains cellulose producing strains and select a variety of protein,fat decomposition microbes mix build composite strains ZW-1,through the liquid and solid fermentation test and microbial diversity detection method to study the degradation characteristics of composite strains ZW-1and its function,composition stability.4)Strains ZW-1function stability: in view of the F10,F15,F20 generation of solid state fermentation product of cellulose enzyme activity of filter paper enzyme activity,the results showed that the compound strains ZW-1 the process of decay,small variations in the composite system of enzyme activity of different generations,each generation of enzyme activity curve changes are basically the same,are presented to reduce trend from low to high,shows that the enzyme production ability of composite has very stable.5)Strains of ZW-1 stability and diversity analysis: usingmicrobial diversity detection method,the rotten compound strains and commercially available bacterium agent ZW-1 A microbial diversity were compared,the results showed that the strains ZW-1the diversity of microbial components.6)In simulation field experiment,the temperature,organic matter as indicators,simulated field covering model has carried on the preliminary field application effect analysis,and measured the p H and seed germination index,indirectly reflects the compound is the growth of bacteria and degradation of performance,for the biodegradable organic material technology research application in improving agricultural environment facilities to provide the basic data.
