Cucumber(Cucumis sativus L.)is a high production and efficiency facility cultivation vegetable crop.“Flowering and acicle” is an important cucumber commodity quality and breeding target traits.Cucumber line ‘6457' can form super ovary,which can delay flowering for 4~5 days and commodity melon is flowering and acicle.It was essential to investigate the development regularity of super ovary in the spring of 2014 and 2015.And it was successful to establish a system of two-dimensional electrophoresis technology based on cucumber female flower's corolla.Furthermore,this could study the super ovary delayed flowering mechanism in proteomics method,aiming to reveal the development regularity of super ovary and delay flowering mechanism.This could accelerate the process of cucumber quality breeding.So far the results were as follows:1.The development regularity of super ovary: The occurrence rate of super ovary was as high as 84.4%,by using cultivation management ways,such as “enough fertilizer and water” “reducing commodity melon load”“timely seedling” “eight days sunlight”.Consequently,adequate sunlight was contributed to form more photosynthetic products,making plants accumulate more photosynthetic products in fruit.In a word,the plants producing super ovary needed strong growth potential and high rate of super ovary occurred in the plant leaf area of 645.5 cm2,the stem diameter of 9.4 mm with continuous sunny days.Conversely,cultivation management ways of normal ovary were “moderate fertilizer and water”“normal mining melon”“normal seedling”,etc.The plants' growth power was middle slightly weak in May.And,the plant average leaf area was 432.6 cm2 and steam diameter was 7.5 mm.So the occurrence rate of normal ovary was 100% with the insufficient sunlight.2.An appropriate two-dimensional electrophoresis proteome analysis system of cucumber female flower's corolla was established by optimizing the parameters including the protein extraction method,the pH gradient of IPG strip,isoelectric focusing conditions,sample application,and dyeing method.This study obtainedclear background,high resolution,and high quality two-dimensional electrophoresis maps,which protein points distributed evenly in phenol extraction method,using 17 cm pH 4-7 IPG strip,as well as optimized isoelectric focusing conditions and silver staining.3.The study of super ovary in proteomics: It was a good way to separate protein in cucumber corolla of super ovary and normal ovary by using two-dimensional electrophoresis.Analyzed by PDQuest 8.0.1.and MALDI-TOF/TOF-MS,73 differentially expressed points was identitied successfully.Formation of super ovary was related with chloroplast of photosynthesis,carbon metabolism,glycolysis energy metabolic pathways.Pyruvate dehydrogenase(PDH2),sulfur,symplectic amide dehydrogenase(LPD1),isocitrate dehydrogenase(IDH1)might be important to further study the formation of super ovary.
