The Hyalomma Asiaticum Morphological Comparison And Gene Polyphism Analysis In Different Regions Chinese

Hyalomma asiaticum is mainly distributed in Asia,and includes 3 subspecies,namely Hyalomma asiaticum asiaticum?H.asiaticum kozlovi and H.asiaticum caucasicum.It is reported that ticks of H.asiaticum asiaticum and H.asiaticum caucasicum often hybridizate in nature,and produce fertile offspring,as these two subspecies have an overlapping distribution in Iran.Therefore,it is difficult to identifiy these two subspecies.Additionally,the two subspecies of H.asiaticum asiaticum and H.asiaticum kozlovi are distributed in China,and coexist in Xinjiang,where can be also found intermediate type species.Thus,it is also difficult to distinguish these subspecies form each other correctly.This study was taken H.asiaticum from different areas of our country as research object,On the basis of the study of Apanaskevich and Horak(2010),the specimens of H.asiaticum have been further analyzed using scanning electron microscopy,digital microscope and molecular biology.Amplified Ribosomal RNA gene of the second internal transcribed spacer 2(ITS2),mitochondrial 16 S r DNA and cytochrome oxidase subunit I(COI)gene sequence of H.asiaticum from 3 provinces of Inner Mongolia,Gansu and Xinjiang including 17 distribution areas.The phylogenetic trees were constructed to analysis the taxonomic status of H.asiaticum,meanwhile,the relative species of H.rufipes and H.anatolicum were compared,and Haemaphysalis qinghaiensis was analyzed as an out group.The results showed that the tick species of H.asiaticum had no distinct geographical isolation,especially ticks from Gansu and Inner Mongolia which were thought to be only occured H.asiaticum kozlovi.H.asiaticum from Gansu,Inner Mongolia and Xinjiang provinces completely mixed together.Thus,subspecies of H.asiaticum kozlovi and H.asiaticum asiaticum should be abolished.Therefore,only species of H.asiaticum occurs in China.
