| In this study,the growth hormone(GH) gene of yellow catfish was cloned,characterized and expressed in Escherichia coil.(1)Molecu Lar characteristics of GH genePrimers were designed according to the conserved region of GH gene and by cloning,sequencing and splicing obtained sequence of GH gene. The sequence analysis resu Lts showed that the fu Ll-length of GH gene was 3117 bp with an ORF(open reading frame) of603 bp that encode a polypeptide of 200 amino acids, with a 5′UTR(5′-untranslated region)of 165 bp and 3′UTR 485 bp, with 5 exons(10 bp, 140 bp, 117 bp, 132 bp and 204 bp respectively) and 4 introns(302 bp, 85 bp, 642 bp and 835 bp respectively).(2)Differential expressionTo compare the expression levels of GH gene in the different tissues of yellow catfish,Real time-PCR analysis was performed. The resu Lts showed that the GH gene was expressed specifically in the pituitary gland, and the expression was not detected in other tissues.(3)Correlation of the SNPs and SSRs of GH gene with growth traitsPolymorphisms of the GH gene were evaluated for associations with growth traits in180 yellow catfish individuals. Using directly sequencing, five single nucleotide polymorphisms(SNPs) and four microsatellite(SSRs) were identified in GH gene, with four SNPs in intron 4(1674G>A, 2100A>G, 2154T>G, 2285T>C), one mutation in 3′UTR(2822A>G) and one SSRs in intron 1(AC) and intron 3(AT), two SSRs in 3′UTR(GA,TCTT). Notably, three SNPs(2100A>G, 2285T>C, 2822A>G) and SSRs(AC, GA, TCTT)were significantly associated with growth traits in yellow catfish. In conclusion, the resu Lts suggesting that these SNPs and SSRs in GH gene cou Ld influence growth traits of yellow catfish and cou Ld be used for marker-assisted selection(MAS) in this species. |
