Early research has shown that Ginseng polysaccharides complex(GSP) effects the immune modulatory of pregnant sows, and influence the growth of the piglets and immune function through the maternal effect. Recently, porcine milk exosomes, found in milk, are the vesicle structure, which contains the immune regulation function mi RNA molecules. This article from the perspective of the porcine milk exosomes, to explore whether GSP by regulating the pig immune related micro RNAs in breast to influence the growth and immune of piglets, analysis the porcine milk exosomes and the piglets serum micro RNAs expression pattern.Random selected the roughly the same period, 90 days during pregnancy health white sows, divided into 2 groups, each group of 10 sows. Using based diet feeding, experimental group is added the 400 mg/kg of GSP in the basic diet, feeding to the end of the weaning piglets. Piglets are weighing within born 24 h and record the farrowing weight and litter weight, as well the number of weak, alive, dead, frozen pigs.on the piglet 14 days of age, recording the piglets weight. Acquisition sow milk of 1st, 3rd, 7th, 14 th day, at the same time, acquisition 1st, 3rd, 7th, 14 th days of blood in front of the piglets vena cava, separation of serum. Using q PCR to detection the immune related mi RNA expression level in control and experimental group of porcine milk exosomes, and ELISA kit in porcine milk immune related indicators, such as Ig G, Ig A, IL-2, IL-6 and so on. Using q PCR to detection the immune related mi RNA expression level in control and experimental group of serum, and the immune indexes in piglets serum was determined by ELISA kit, such as Ig G, Ig A, IL-2, IL-6 and so on.The results showed that:(1) the GSP has no significant influence(p>0.05)on the number of weak, alive, dead, frozen piglet. Statistical analysis showed that GSP have a certain role in promoting the growth of the piglets, the piglets weight in experimental group of 14 th days is significantly higher than control group(p<0.05).(2) GSP can significantly improve the Ig A expression levels(p<0.05) of sow lactation at the 3rd day, significantly improve the Ig G, IL-2, IL-6 levels of sow lactation at the 1st and 3rd day(p<0.05). Improve the expression level of serum Ig A at the 3rd day after piglets born(p<0.05), significantly improve piglets 1st and 3rd day of serum Ig G, IL-2, IL-6 expression levels(p<0.05).with linear regression analysis, we found that the sow milk and piglets serum Ig G, Ig A expression levels were positively correlated(p<0.05) and maternal effect is remarkable.(3) using fluorescence quantitative analysis, we found that porcine milk exosomes immune related mi RNA expression level began to increase at 1st day, the 3rd day to reach the highest, and no changes at the 7th and 14 th days, the piglets serum immune relevant mi RNA expression level reach the highest at the 1st day, is lower in the 3rd day, and the 7th day and 14 th day are basically remained stable. Besides mi R-30a-5p, the expression level of porcine milk exosomes mi RNAs is smaller than that in the piglets serum, about10 to 100 times.(4) compare the immune relevant mi RNA of porcine milk exosomes and piglets serum, we found that GSP has different regulation on them. Lactation of the 3rd day, mi R-193a-3p, mi R-320, mi R-30a-5p were significantly lower than that of control group(p<0.05). on 1st, 7th, 14 th days are also has a tendency to lower, the expression levels of mi R-92 a and mi R-140-3p in porcine milk exosomes and piglets serum has obviously raised, while the mi R-92 a was up significantly(p<0.05), on 1st, 7th, 14 th days are also has a tendency to up-regulation.This study show that GSP can regulate the immune related micro RNAs expression of porcine milk exosomes, and the exosomes can through the intestinal canal be absorbed by piglets.
