The chronic poisoning diseases of AFB1 occur frequently, as to this problem, researches on use of chinese medicine to prevent are gradually further. This subject mainly observe the pharmacological effects of "Compound Chai Qin particles" on liver injury and discuss the effects of "Compound Chai Qin particles" for the activity of cytochrome P450 enzymes, mRNA and protein expression on molecular level when getting liver injury, which could provide theoretical basis for clinical safety use and combined use of "Compound Chai Qin particles".The main test content and results are as follows:The experimental methods126 One-day-old ducklings,feeding trials based diet for 6 days.21 seven-day-old ducklings were made as normal control group.125 seven-day-old ducklings were feed trials based diet with 40?g/kg AFB1 to set up AFB1 chronic poisoning model of duckling and they were divided into five groups, including model group, positive control group, Compound Chai Qin Particles high-dose group, Compound Chai Qin Particles middle-dose group, Compound Chai Qin Particles low-dose group,21 in each group. After injury 7,14, 21 day, the serum was taken to detect of AKP activity, GOT activity, GPT activity, SOD activity and MDA content.2g liver to produce tissues livermicrosomes were taken to detect of the activities and protein expression of CYP3A4, CYP1A2 and CYP2E1. The real-time fluorescence quantitative PCR were used to investigate the influence of "Compound Chai Qin Particles" on the mRNA expression expression of CYP3A4, CYP1A2 and CYP2E1.The experimental resultsCompared with model group, AKP activity, GOT activity and GPT activity was also decreased significantly (P<0.05), SOD activity increased significantly, but MDA content significantly decreased in serum of the three dose groups (P?0.05). Liver vacuolation and bile duct proliferation weakened. And dosing for 14 days, the indexes were closer to normal.Compared with model group, activities of CYP3A4, CYP1A2 and CYP2E1 in liver was decreased significantly in three dose groups on 7th and 14th day(P<0.05). The activities of CYP3A4, CYP1A2 and CYP2E1 was increased in three dose groups on 21th day but no significant difference were realized between model group and Compound Chai Qin Particles groups (P<0.05).The relative expressions of mRNA and protein of CYP3A4, CYP1A2 and CYP2E1 in groups with different doses were significantly lower than those of model group in this experiment (P<0.05). The mRNA expressions and protein expressions of CYP3A4, CYP1A2 and CYP2E1 of the three dose groups on 7th and 14th day were significantly lower than those on 21th day (P<0.05). And the data of low-dose is significant higher than high-does and middle-dose (P<0.05).ConclusionCompound Chai Qin Particles could effectively control duck's chronic poisoning of AFB1, by regulating the antioxidant and strengthening liver function. And the best therapeutic regimen is 14 days with low-dose.Compound Chai Qin Particles could effectively inhibit activities of CYP1A2, CYP2E1 and CYP3A4 in early treatment phase. This may be due to decrease of activation of AFB1 and that reduce chemical liver injury to attain the role of protecting liver. The activities of CYP450 were increased on 21th day.Those may be caused by Compound Chai Qin Particles which could increase activity of CYP450,with time going, but its induction is far less than AFBTs.Compound Chai Qin Particles could effectively decrease mRNA expression and protein expression of CYP1A2, CYP2E1 and CYP3A4 in liver microsomes of AFB1 chronic poisoning ducklings and protect the liver. This may be due to decrease of activation of AFB1 and that reduce chemical liver injury to attain the role of protecting liver. However as the medication time of growth, the mRNA expression and protein expression of CYP1A2, CYP2E1 and CYP3A4 has gradually increased, Those may be caused by Compound Chai Qin Particles which could induce of CYP450,with time going, but its induction is far less than AFB1's.
