The Study Of Milbemycin Oxime On Safety In Collies And Curative Effect Of Canine Scabies And Demodicosis

Milbemycin oxime is an antibiotics,produced by Streptomyces hygroscopicus through fermentation.The main mechanism of Milbemycin oxime is connect with specific pots in the body of worms and other arthropods,when can cause a series of biochemical,physiological reactions,and death.Since its discovery,Milbemycin oxime is widely used as exterior and interior anthelminthic in dogs and cats.In this study,the safety of Milbemycin oxime on collies,its clinical effects on the treatment of the natural infectionwith canine scabies mites,and demodico would be explored and evaluated,and then was provided guidance for its application in the pets clinical.Test Ⅰ Safety of Milbemycin oxime in colliesThe test was conducted to evaluate the safety of Milbemycin oxime in collies,twelve healthy adult collies about 15kg were selected and randomly divided into four groups of three.According to the recommended clinical dose of the drug(0.5 mg·kg-1),they were set 0 time the recommended clinical dosage groupA(0 mg·kg-1).the recommended clinical dosage group B(0.5 mg·kg-1),3 times the recommended clinical dosage groupC(1.5 mg·kg-1),5 times the recommended clinical dosage groupD(2.5 mg·kg-1).Respectively,they were administered once daily and last for 4 months.During the test,dogs’ clinical manifestations were observed.Hematological,blood biochemical,urinalysis and Pathological changes were compared on the day before the test,4,8,12 and 16 weeks.The results showed that the hematological indexes had no significant difference between group A and group B、C、D(P>0.05).The ALT,AST,TBil,TP,BUN,CREA and other blood biochemical indexes were in the normal range.Test Ⅱ C linicalefficacy of Milbem ycin oxime in canine on S.scabies mitesTo exPlore the theraPeutic effect of Milbemycin oxime in canine on scabies mites,27 natural infection with scabies mites canines were selected and randomly divided into 5 groups including blank control group A without medicine(n=3).The remain 4 groups was set as:the Ivermectin group B(0.5 mg·kg-1),the recommended clinical dosage group C(0.5 mg·kg-1),3 timestherecommended clinical dosage group D(1.5mg·kg-1),and 5 times the recommended clinical dosage group E(2.5 mg·kg-1).the test was last for 4 weeks.The mites in skin was observed by the scrape method,at the same time,the clinical symptoms,SSLS and GCS were scored to evaluate the treatment effect of Milbemycin oxime on scabies mites.The results were showed that,There was no significant difference among group B、D、E(P>0.05).Al1 dogs were cured,and the SSLS,GCS score were normal.However.there were significant difference between group C and group A and group B、D、E(P<0.05).The results proved that,1.5 to 2.5 mg·kg-1 dose,once a week,for three weeks in a row,could be effective on scabies mites,and that the clinical symptoms,very highly security.Test Ⅲ C linicalefficacy of Milbem ycin oxime in canine with demodicosisTo explore the therapeutic effect of Milbemycin oxime in canine with D.demodex,15 natural infection with demodicosis canines were selected and randomly divided into 5 groups of 3,the group were:ivermectin group A,the recommended clinical dosage group B(0.5 mg·kg-1),3 times the recommended clinical dosage group C(1.5mg·kg-1),5 times the recommended clinical dosage group D(2.5 mg·kg-1).Administered once daily.Dogs which clinical symptoms disappeared and skin microscopy inspection found no mite in consecutive 2 months were regarded as cured and the drug were removed.The whole test cycle lasts for 4 monthes,after 4 monthes,if can still see the mites,were treated as failure.The mites in skin was observed by the scrape method,at the same time,the clinical symptoms,SSLS and GCS were scored to evaluate the treatment effect of Milbemycin oxime on demodicosis.The results were showed that,the descent rate of demodxe were about 100%and cure rate above 67%,in the treatment of the second month,some dogs in group C、D were cured.Meanwhile all dog in group A were cured in the third or fourth months.The results proved that,1.5 to 2.5 mg·kg-1 dose,once daily for two monthes in a row,can be 100%reduce the quantity of demodicosis,and obviously improve the clinical symptoms,very highly security.
