In this study we constructed the oxidative stress model of chicken skeletal muscle cells to explore the mechanism of adiponectin in the oxidative damage of chicken skeletal muscle cells with the participation of p53 lysosome and mitochondria.It provided the basis for broiler's anti-stress and efficient breeding.The results are as follows:1 Culture and identification of skeletal muscle satellite cells were established,and the oxidative stress model was established by hydrogen peroxide treatment.The results showed that:(1)the DESMIN cell culture immunofluorescence for identification of skeletal muscle satellite cells;(2)hydrogen peroxide treated cells,ROS and MDA levels and TUNEL staining significantly increased the rate of cell apoptosis,cell activity,SOD and GSH-Px enzyme activity unit decreased significantly(P<0.05).Oxidative stress model of skeletal muscle satellite cells treated with hydrogen peroxide successfully.2 Adiponectin treatment of oxidative stress cells,analysis of related indicators of lysosomal and mitochondrion.The results showed that:(1)ROS,MDA level and cell apoptosis rate of TUNEL in adiponectin treatment group was significantly decreased,cell activity,SOD and GSH-Px enzyme activity unit increased significantly(P<0.05);(2)adiponectin treatment group significantly improved the permeability of lysosomal and mitochondrion membrane,Bax,Cyt-c,Caspase 8,Cathepsin B,Chymosin gene and protein level were significantly decreased,Bcl-2 mRNA and protein levels were significantly increased(P<0.05).Adiponectin repair cell damage induced by oxidative stress via lysosomal and mitochondrion.3 Study on the mechanism of p53 involved in the repair of oxidative stress by adiponectin treatment with p53 inhibitor PFT-?.The results showed that: Compared with the APN treatment of oxidative stress in the model group,(1)PFT-? further significantly reduced the cell ROS and MDA levels and increased the apoptosis rate of TUNEL,cell activity,SOD and GSH-Px enzyme activity unit(P<0.05);(2)further improved the membrane permeability of lysosomes and mitochondria,Bax,Caspase 8,Cyt-c,Cathepsin B,Chymosin gene and protein level were significantly decreased,Bcl-2 gene and protein levels were significantly increased(P<0.05).In conclusion,adiponectin can inhibit the expression of p53 to improve the lysosomal and mitochondrion membrane permeabilization,restoring the membrane potential,inhibiting the expression of apoptosis gene and protein,promoting the expression of anti apoptosis gene and protein,inhibiting the apoptosis process and cell injury caused by oxidative stress.
