| In this study, new varieties of Amorphophallus "Chu Mo Hua No.1" as the research object, tubers, petioles, leaves and shoots as explants tissue culture when looking for the best performing tissue culture inoculation material. At the same combinations of different concentrations of hormones, the most suitable for the induction of callus induction, bud differentiation and proliferation rooting medium components, the establishment of "Chu Mo Hua No.1" rapid propagation system. It laid the foundation for the next Amorphophallus tissue culture factory production.Now we get the following results:1. Konjac tuber using as explants, after four different methods of disinfection sterilization treatment, were plated on MS+1.0mg·L-1 NAA+1.0mg · L-16-BA medium. The results showed that the best sterilization method:first, using 0.1% Tween-20 solution concussion 8min, then using 75% alcohol soaked 30s, sterile water and then washed three times, each rinse 1min, and finally using 0.1% HgCl2 12min soaking solution, sterile water five times rinse lmin.2. Konjac tuber as explants, after disinfection were cut in different types, concentrations of anti-oxidants solution and then unified seeded on MS+lmg · L-1NAA+1mg·L-6-BA. The results are:cutting explants when, at 0.5g/L of Na2SO3 solution was carried out to reduce the browning when tissue culture explants browning rate of 50%.3. Different explants konjac after sterilization treatment, were plated on MS+lmg· L-1NAA+lmg· L-16-BA. Induced the highest rate of bud explants, induction rate reached 78.26%, is the best in this experiment explants.4. The konjac tuber as explants, primary culture, the most suitable konjac callus induction medium was MS+1.5mg·L-1 NAA+1.5mg·L-16-BA, induction rate 83.62%.5. The konjac tuber as explants, the most suitable medium is MS+0. Olmg· L-1 NAA +2.0mg·L-16-BA of adventitious buds induced, induction rate 96.15%, value-added factor was3.1.6. The konjac tuber as explants, the more suitable medium is:MS+0.5mg·L-1 NAA of the rooting medium, the rooting rate 94%, the average root number 4.6, the longest root length 6.2cm.7. The robust plantlets transplanting after acclimatization, using humus and perlite geometric 1:1 formulation matrix, konjac plantlets highest survival rate reached 73.33%. |
