This study is focusing on rabbit, using low-fiber diet to induce intestinal disorder. The experimental individuals are divided into healthy, mild and severe three groups according to disease severity. By using qRT-PCR detect enteritis-related 25 mRNA and 25 miRNA in ileum, colon and sacculus's different expression in three different groups, revealing digestive disorder's variation tendency under the circumstance of varied inflammation degree from the transcript level, as well as disorder of digestive system's variation in ileum, colon and sacculus. Furthermore, we performed correlation analysis between mRNA and miRNAs to uncover cooperative roles in rabbit nonspecific digestive disorder process's regulatory functions. The research results are as follows:1. Analysis results on mRNA shows that expression of JAK2, IRGM and IFN-?in ileum was significantly higher than sacculus and colon (.P<0.05); expression of IL-10. IL-22, TNF and IL-1B in sacculus was significantly higher than ileum and colon (P<0.05); expression of S1AH2, IkBa, IL-18, NP-5, DEFB135 and NUMB in colon was significantly higher than sacculus and ileum (P?0.05); as the nonspecific digestive disorder getting more severe, the expression of ZEB1, SMAD2, NP-5. TLR4, NOD2, NLRP3, JAK1, JAK2, STAT3, TNF, IFN-?, IL-1B, IL-18 and IL-22 increased, while the expression of RHOB. SOCS-1, ATG5, ATG16LJ, IRGM, DEFB135, SIAH2, NLRPI2, IkBa, NUMB and IL-10 decreased.2. Analysis results on mRNA shows that the expression of miR-31, miR-29a and miR-151-5p, miR-106a, miR-106b, miR-195, miR-30c, miR-130a, miR-146b, miR-132, miR-141, miR-29b and miR-192 in ileum were significantly higher than sacculus and colon (P?0.05); the expression of miR-155 and miR-142-3p in sacculus was significantly higher than ileum and colon (P<0.05); the expression of miR-199-5p. miR-10a. miR-122, miR-126 and miR-200b in colon were significantly higher than sacculus and ileum (P<0.05); as the nonspecific digestive disorder getting more severe, the expression of miR-21. miR-126. miR-29a, miR-31, miR-151-5p. miR-199-5p. miR-106a. miR-106b, miR-142-3p, miR-195. miR-362-3p, miR-30c, miR-130a, miR-155. miR-146. miR-146b, miR-132, miR-150 and miR-14] increased; while the expression of miR-29b. miR-192, miR-122. miR-10. miR-124. miR-124 and miR-200b decreased.3. In these three different kinds of intestinal tissues, there are 113 pairs of miRNA-mRNA that hold significant negative correlation, in which 15 pairs of them have specific target relationship according to published articles. The results show that the variety of protein-coding genes and miRNA participate in the process of regulating non-specific digestive disorders, and show the different expression profiles, lay a foundation for further study of non-specific digestive disorders mechanism, and accumulated basic data for IBD study using rabbit as an animal model.
