China's tobacco planting and production quantity ranked first in the world?It is an urgent need to study and solve the problem of waste tobacco rod in the process of tobacco harvest and transportation. The purpose of this experiment is to change the waste tobacco rod produced in cigarette manufacturing into the needed organic fertilizer in agricultural production, and determine the optimal compound bacteria on the basis of the previous research in this laboratory. Will the compound bacteria to control tobacco stalk solid fermentation test, to observe the dynamic changes of its fermentation process, analyzed the main factors that affect the organic matter degradation, solid state fermentation conditions optimization, define and verify the best solid fermentation conditions. The main results are as follows:1:By tobacco stalk selective medium to get the final screening, determination of cellulase activity and strain pairwise antagonistic test, no mutual obvious antagonistic and cellulase activity of high strain H2568, B1, M1 and N1. They are three strains combination and combination of four strains of tobacco rod liquid fermentation test, to determine the high temperature resistance of effective compound bacteria H2568-B1-M1. The activity of the enzyme reached the highest 625.44 U/ml, strain H2568, B1 and M1 to 1:1:1.5 ratio fermentation 15 days, the cellulose degradation rate, hemicellulose degradation rate, lignin degradation rate reached 56%,70%,33%, the degradation degree of organic matter and reducing sugar content is missed bacteria of tobacco rod has greatly improved.2:Composite bacteria without regulation of tobacco stalk solid fermentation test results show:compared with the liquid fermentation, the highest cellulase activity decreased to 213.29 U/ml, fermentation substrate in cellulose degradation rate was 38.91%, hemicellulose degradation rate was 49.24%, lignin degradation rate for 24.43% total nitrogen content changed little, reducing sugar content from 5.92mg/g before fermentation to the 21 days to 47.37mg/g and water rate in 44% to 62% fluctuation. Compared with liquid fermentation of tobacco rod, temperature, moisture, pH may be the main reason of fermentation substrate organic matter degradation rate.3:Through the single factor test, inorganic salt ions and smoke rod solid-state fermentation conditions optimization test show that the optimal conditions for:pile turning frequency for 7 days, the best expanding loose agent for 10% of the straw, add 0.30%PO42+, 0.20%Mg2+ and 0.10%Ca2+ three kinds of inorganic salt ions and initial water content rate of 50%,21 days of fermentation,8% inoculation quantity, initial pH5. The results showed that the degradation rate of cellulose, hemicellulose and lignin reached 59.15%,75.20% and 36.44% respectively, and the content of reducing sugar reached93.21mg/g, which was basically in line with the expectation.
