In recent years,with the sequencing of grape genome and the release of genome sequence data,the molecular biology and functional genomics in grape has become an important research topic.However,flower development in grape is a complex biological process,and the related research is only at a preliminary stage,particularly in China,the related research not only far behind the model plant Arabidopsis,but also significantly behind other important fruit such as pears,apple.In this study were selected grape cultivar'Summer Black','Fujiminori','Drunk Jinxiang' as materials to explore the impact of hormone treatment and cultivation measures on the flower and fruit development from the perspective of molecular biology and physiology respectively.The main findings are as follows:1.This study focused on the influence of different branch drawing treatments on flower development-related gene expression during the flowering process of grape winter buds.Using 'Summer Black' grapes(Vitis vinifera × V.labrusca 'Summer Black')as test materials,the semi-quantitation RT-PCR was applied to analyze the expression of five important floral genes,including VvFT?VvSOC1?VvLFY?VvAP2 and VvFLC,by different drawing treatments to the winter flower buds on the long,medium,short branches of'Summer Black' grapes.The result showed that different treatments have differences in the expression patterns.The expression of level branches is the highest.In the same drawing treatment,there are also differences among the long,medium,short branches,and the expressions of the medium,short branches are obviously higher than the long one.2.This experiment used grapevine(Vitis vinifera L.'Fujiminori')as test materials,the fluorescence quantitative RT-PCR were applied to analyse the gene expression in grape pollen abortion of miR159 and its target GAMYB-like gene under Gibberellin mobilizing during the different development period.The result showed that the best treatment was 8-days postbloom,followed by 4-days and 12-days postbloom.This research indicated that the molecular mechanisms of Vv-miR159 response gibberellin in regulation the grape pollen abortion.3.This experiment used grapevine('Drunk Jinxiang')as test materials,with the treatment of three different concentrations of GA(20ppm,40ppm,60ppm)and auxin(lOppm,20ppm,30ppm)during about a week before flowering,study the effects of different concentration of GA and auxin on the length of flowers,the length of fruit spike internode and fruit aspect ratio and the expression level of flower development related gene.The results show that:there are significant differences in expression level of genes related flower development between different concentration of gibberellin and auxin,and 40ppm concentration of GA3 treatment had the best effect on promoting flower development.
