The Effect Of Different Adhesion?Fixed Liquid And Frost Crystal Method On The Quality Of Frozen Slices

Frozen section technique is a kind of commonly used veterinary pathology inspection technology,mainly used in veterinary clinical rapid diagnosis,dyeing and fat tissues antigen-antibody reaction or enzyme detection.But in veterinary clinical pathology diagnosis,often appear due to poor quality of frozen section lapse in judgment.In this experiment to improve the quality of frozen section,animals' heart,liver,spleen,lungs,kidneys and muscles as the experimental material,such as using conventional histologic and frozen section technique,different adhesive,stationary liquid and the ice crystals method to research on the effects of the quality of frozen section.The results showed that:1.The effect of different adhesion on the quality of frozen slices in fixed tissue.By protein,glycerin,poly lysine,three kinds of adhesive gelatin chromium potassium sulfate treated glass slide and purchase the slide of dropping from reagent company effect exists obvious difference.Treated by more poly lysine slides to take off the piece rate was 0%,the resistance effect is best,followed by gelatin chromium potassium sulfate(at a rate of 5%),and the protein glycerin processed glass slide,and purchase of slide off from reagent company rate was 66%and 92%,the resistance effect is poorer.2.The effect of different fixed fluid on the quality of frozen slices of fresh tissue.The effect of six types of fixed fluid on the production of frozen slices of fresh tissue and the effect of H.E dyeing were different.The effects of ethyl ether alcohol 1:1 were the most effective,followed by AAF fixed fluid,4%polyformaldehyde,and 95%ethanol,medium follicin and AF fixed fluid.3.The effect of different ice crystals on the quality of frozen slices of fresh tissue.Different ice prevention method were to ice crystals,but eliminate the level differences,including liquid nitrogen cooling method and isopentane directly handle 25 s of isopentane-liquid nitrogen method the effect is best,and sucrose gradient encapsulation-dehydration and isopentane processing 20 s and 30 s of isopentane-liquid nitrogen method results are poor.Therefore,making a high quality of frozen section,should adopt the following methods:fixed organization frozen section,using poly lysine processed glass slide;When making fresh tissue frozen slices,ethyl ether alcohol 1:1 fixed solution and liquid nitrogen direct cooling or isopentane treatment of 25 s of isopentane and liquid nitrogen to prevent ice crystals.
