As the average annual moth-eaten rate of the soybean pod borer,Leguminivora glycinivorella(Matsumura),is 10%-15%,seriously affecting the quality and yield of soybean,soybean pod borer is an important pest of soybean.The mature larvae of soybean pod borer spend 10 months diapauseing in cocoon,only one generation each year,so the study of diapause mechanism can provide evidence for the monitor and control of soybean pod borer.At present,there are only some ecological studies on the diapause of soybean pod borer,and physiological of diapause of soybean pod borer have not been reported.This paper studied the effects of host plants,temperature of diapause induction period,temperature of diapause period and diapause time on the biochemical substances content of soybean pod borer from diapause to pupae,in order to predict the diapause development stage and the number of effective surviving populations after relieving diapause,and provide the basis for monitoring and control of soybean pod borer.In this study,we determined the contents of biochemical substances such as fat,water,carbohydrate,protein and glycerol of soybean pod borers fed on soybean and wild soybean,and the soybean pod borers set at temperatures of 17?,20 ?,23 ? and 25 ? in diapause induction period,and the soybean pod borers was set at temperatures of 0?,17? and 25?in the diapause stage,and soybean pod borers of the different diapause stages,and the soybean pod borers in outdoor,by anthrone-sulfuric acid method,methanol-chloroform extraction method,acetylacetone colorimetric method and Coomassie brilliant blue G-250 staining method.The effects of temperature,nutritional conditions and diapauses depth on the diapause of soybean pod borer were clarified The main results of study were as follows:We measured and compared the contents of biochemical substances of soybean pod borers fed on cultivated soybean and wild soybean during diapauses,and the results showed that the host of soybean pod borer has the impact on the content of biochemical substances.The contents ofprotein and total sugar of soybean pod borer fed on wild soybean was significantly higher than that fed on cultivated soybeans,while the content of water was lower than that of cultivated soybeans.We measured and compared the biochemical substances content of soybean pod borer in diapause induction period set at different temperatures,and the results showed the content of fat,water content,carbohydrate and protein with different treatments was not related to temperatures in diapause induction period,and the content of glycerol could be influence to temperatures in diapause induction period and decreased with the increase of temperature in diapause induction period,especially the content of glycerol of soybean pod borer at 25 ? was 2.38?g/mg significantly lower than other temperature.The soybean pod borer diapausing at different temperature could relieve diapauses,and some soybean pod borer at 17 ? and 25 ? were earlier than at 0 ?.It is found that the content of biochemical substances of soybean pod borer in outdoor conditions were similar to the low temperature(0?).The content of trehalose and glycerol of soybean pod borer in low temperature was significantly higher than that in middle and high temperature condition in the middle days of diapauses;The contents of total sugar,glycogen and protein of soybean pod borer in middle temperature were higher than other condition;The water content of soybean pod borer in high temperature was higher;the content of fat of soybean pod borer would be lower with diapauses temperature higher at the end of diapauses.The content of carbohydrate,protein,fat and glycerol of soybean pod borer has increased significantly,while the water content of insects decreased entering into diapauses stage.The analysis of biochemical substances changes in the whole diapause stage showed that January and February of winter were an important turning point in the change of biochemical substances of soybean pod borer,and the content of glycerol and trehalose reached the maximum at 4.78?g/mg and 69.81?g/mg,and total sugar,glycogen and protein content decreased,and fat content increased.The content of glycogen decreased and the content of trehalose increased in the whole diapauses stage.Indicating that the changes of glycogen and trehalose showed a mutual transformation of the relationship,the soybean pod borer were the trehalose accumulation type.The content of glycerol was negatively correlated with the temperature,the lower the temperature,the higher the glycerol content.
