Viral Replication And Apoptosis Of MDBK Cells Infected With BVDV And IBRV

Bovine infectious laryngotracheitis virus(IBRV)and bovine viral diarrhea virus(BVDV)are two important infectious diseases of cattle.Two viruses can cause immune suppression.BVDV and IBRV co infection,BVDV and other respiratory pathogens mixed infection or secondary infection.In this study,we investigated the viral replication and apoptosis of two kinds of virus infected MDBK cells in vitro.This study first conducted BVDV and IBRV type CP proliferation,and then select 0.2 MOI,1 MOI,2 MOI BVDV infection of MDBK cells,50% cells were infected with IBRV lesions(0.1 MOI),the control group and the blank cells and separate the infection of BVDV,IBRV control.Co infection of 24 h cells were collected after extraction of RNA by real-time fluorescent quantitative PCR and indirect immunofluorescence method of virus infection,detection of load and cell activity,identified two kinds of virus replication and cell activity after virus infected MDBK cells.CCK-8 cell proliferation assay,TUNEL assay and flow cytometry were used to detect the location and apoptosis of the virus,and to determine the effect of CO infection on apoptosis.The results showed that when BVDV 0.2MOI and 1.0MOI,the number of copies of IBRV was not significantly different from that of IBRV alone.When BVDV was 2 MOI,the copy number of IBRV virus was significantly lower than that of IBRV alone.Indirect immunofluorescence assay showed that BVDV and IBRV could infect MDBK cells and induce apoptosis,and with the increase of the amount of BVDV virus,the pathological changes of cells increased.Cell activity test results showed that the co infection of two viruses of cells caused a greater impact,with the increase of the amount of virus,cell activity decreased,and the common infection group was lower than that of group cell activity in cells infected with BVDV alone.The results of flow cytometry showed that the infection rate of CO infection with BVDV and IBRV was higher than that of BVDV infected cells,and the effect of BVDV on the replication of IBRV was not significant.In summary,research shows that BVDV infected MDBK cells infected IBRV,two virus can replicate in cells,but high multiplicity of infection BVDV will increase MDBK cell lesions,thereby affecting IBRV replication.The co infection of BVDV and IBRV decreased the activity of the cells,and the damage to the cells was greater than that of the virus,which caused the cell apoptosis.
