Phosphorus is involved in the synthesis of ATP and plays important role in multiple enzyme activity that related to latex production.So,Pi can measure the rubber production potential.The more latex is producing,the more phosphorus is running off.To ensure the constant production of latex,the rubber tree must recover its phosphorus inside as soon as possible.In plants,Purple acid phosphatase(PAP)can degrade phosphorus storage,then release inorganic phosphorus,and finally promote the reutilization of phosphorus.The phosphorus transporter is responsible for the absorption and transport of phosphorus.Up to now,research on phosphorus transporter and purple acid phosphatasein in rubber trees is blank.Therefore,we used two experimental materials,PR107 and RY7-33-97 latex,were stimulated by ethylene,the control group were not.Then we proceed correlation analysis for total phosphorus,inorganic total phosphorus,B-serum inorganic phosphorus,C-serum inorganic phosphorus and its expression of phosphorus transporter gene and purple acid phosphatasein in a ethylene stimulating cycle.Through this analysis,we can study the effect of ethylene-stimulated dicing on inorganic phosphorus metabolism in rubber latex,the results are as follow:1.Physiological IndicatorsThrough the test of rubber trees’ sucrose,soluble protein,thiol,we found those indicators level were both higher than the control group after PR107 stimulated by ethylene,which means PR107’s stress and stimulation resistance enhanced.RY7-33-97 sucrose supply was not enough,soluble protein content decreased,mercaptan content was relatively stable,the thiol content was stable,all those indicated that RY7-33-97’s stress and stimulation resistance decreased after the ethylene stimulation.With the increase of cutting times,inorganic phosphorus and phosphorus content of rubber tree has a trend of slow recovery followed with a distinct increase.The inorganic phosphorus and phosphorus content of ethylene treated group was much lower than no-ethylene treated group.The inorganic phosphorus content of ethylene treated PR107 was much lower than RY7-33-97.With the increase of cutting times,inorganic phosphorus of B-serum and C-serum in rubber tree has a trend of slow recovery followed with a distinct increase.The overall change trend of inorganic phosphorus content of PR107 was similar to RY7-33-97,which means B-serum inorganic phosphorus content Oh>1h,C-serum inorganic phosphorus content was the opposite of this.Through the test of the nucleic acid of the rubber tree,we find an overall rise in DNA content and RNA content and showed an accumulation trend.The ratio of RNA/DNA was increasing in the first cut and the fourth cut.The increase of the first cut was the most significant.Transcription and translation function gradually increased,protein and ribosomes gradually accumulated,which are conducive to gene expression.2.The correlation between Physiological Indicators and PT,PAP expressionThe stimulation of ethylene can rise the expression of Hbphtl:1 gene.There is a distinct relation between physiological indicators and gene expression.With the loss of phosphorus,up-regulation of Hbphtl:1 expression inhibited phosphorus running off excessively,which PR107 was higher than RY7-33-97 in this progress.The decrease of inorganic phosphorus content was significantly lower than RY7-33-97,because the inhibition of phosphorus loss on PR107 was stronger.The expression of HbPAP18 gene was up-regulated by ethylene stimulation.With lossing phosphorus,the phosphorylation activity of HbPAP18 gene was rised,activating phosphorylase restored,then the phosphorus pool degraded in the lutoid so as to add deficiency of phosphorus,which the degradation was higher than control group.The stimulation of HbPAP26 gene down-regulates the expression of ethylene,which was closely related to the content of inorganic phosphorus in plants.When the content of inorganic phosphorus was high,the effect of down-regulation was obvious,and the effect of down-regulation was not.The effect of ethylene on the PAP and PT regulation of rubber tree had significant characteristics:PR107,the low yield resistant strain,was significantly up-regulated by HbPAP18 and down-regulated slightly by HbPAP26 after ethylene stimulated,which promoting decomposing organophosphorus in plant.There was a some trend of decline in inorganic phosphorus of PR107 in each knife.3.Subcellular LocalizationThrough constructing pBWA(V)HS-Hbphtl:l-GLosgfp and empty vector to transfer into rice protoplast cells.The expression of the plasmid was observed under a laser scanning confocal microscope,the result showed that Hbphtl:1was located on cell membrane.
