Rapeseed,one of the most efficient oil crops in China,is the best choice to ensure and improve the self-sufficiency rate of edible oil in China.Nowadays,rapeseed production in our country is developing rapidly with high oil content,high-yielding and high efficiency.Heterosis is an important way to increase yield per unit area of rapeseed,and nuclear sterility is one of the effective ways for rape heterosis utilization.9012 AB is a representative type of nuclear male sterile.Its genetic model has been studied clearly.With the three-line breeding method of this system,a number of fine varieties have been developed.In this research,based on the latest research results of 9012 AB system,we used high oleic acid molecular markers which had been developed in our laboratory to breed the genic male sterile line and its all maintainer line with high oleic acid in Brassica napus L.and we combined backcross with molecular marker-assisted selection in the breeding.The main results are as follows:1.Based on the difference between BnMs3 and Bnms3 sequences,allele specific markers GQP 209F/GQP 209 R were designed.The new marker has a difference of 14 bp between the size of the amplified products of the two pairs,which can distinguish between the two sequences.The new markers were applied to the materials with different genetic backgrounds,and the polymorphism was found to be different.2.5 markers about BnMs4 loci in the literature were evaluated.Co dominant marker P4 is not suitable for materials in this study;the detection effect of co dominant marker ZHP 14F/R by agrose electrophoresis is not ideal;the detection effects of dominatnt marker S25 and ZHP 9F/R are good;Co dominant marker A7-24 F/R is suitable for each generation of materials with different genetic background,and it is high efficiency.3.To assist the selection by molecular genetic markers and molecular markers of high oleic acid,BnMs3 and BnMs4 loci of each allel are induced into ZH5,ZH5-1,ZS9 and MS1-D which are the recurrent parent in excellent.Genotype identification,florescence fertility investigation and determination of oleic acid and oleic acid analysis showed that the fertility of genotype and phenotype are basically the same.It proves molecular markers can quickly integrate high oleic acid and fertility genes into high-yield varieties(lines).4.Primers of AFLP were used in the background selection of different sources of BC1F1,BC2F1 and BC2F2.We got BC1F1 double heterozygous strains which are high PRPG and BC2F2 double heterozygous male sterile plants,maintainer lines and all maintaine lines.PRPG in BC1F1,BC2F1 and BC2F2 are 58.33%-94.44%,81.58%-100.00% and 82.35%-96.36%,respectively,which provide important information for selecting high background recovery materials.5.We obtained 10 homozygous double purpose lines(AB)and 16 all maintainer lines(C)with high oleic acid gene through molecular marker assisted selection in the BC2F2 population of ZH5,ZH5-1 and ZS9.
