Abstract/Summary:
Walnut(Juglans regia)is a walnut walnut is a deciduous fruit trees,belonging to perennial economic trees.As one of the world’s four major nuts,its core has a very rich nutrition such as rich in high quality protein,a variety of mineral elements and other nutrients,high fat content,known as "the king of wood oil" title.Walnut planting area is expanding,China has become the world’s largest growing country,walnut acreage in 2012 about 425,000 hectares,the output of about 1.7 million tons(FAO),but in recent years,environmental changes and the impact of pests and diseases Walnut production and economic benefits caused by the huge losses,of which walnut bacterial black spot disease is caused by bacterial infection,also known as black spot disease,the disease is widely distributed in all major walnut producing areas around the world,serious harm Walnut production,disease-related protein(PRP)is an important disease-resistant response protein,but rarely reported in walnuts.In this study,PR1 and PR4 genes were cloned from walnut by using "Hongling" walnut.The PR1 and PR4 genes were compared by bioinformatics analysis.The real-time fluorescence quantitative technique was used to compare the two genes in different walnuts The expression level of PR1 and PR4 in the pathogen of bacterial black spot was analyzed.The main results of this study are as follows:(1)The PR1 and PR4 genes of walnut were cloned by T-A and sequenced.The sequence characteristics of these proteins were analyzed by BLAST,evolution analysis and multiple order comparison.The PR1 gene of walnut was homologous to the PR1 gene of rice and wheat,and the PR4 gene of walnut was homologous to the PR4 gene of grape.(2)walnut bacterial black spot serious damage to walnut production,bacteria in the winter latent in the walnut winter buds,PR1 and PR4 gene in different walnut varieties in the winter buds analysis found that PR1 in the West 1,Liao 3,Xiangling and Lin 3 were expressed in the expression of winter buds,while the expression level in the phenanthrene and Liao 4 was lower than that in the control.The PR4 was in the order of Xipu 1,Liao 3,Hong Ling,Lin 3 and Qi The results showed that the expression of PR1 and the gene of different walnut varieties were different,and the resistance of different walnut varieties was predicted to be different.(3)Inoculation of the pathogen of bacterial black spot was carried out by using acupuncture inoculation method.The results showed that PR1 and PR4 could be used for different varieties of walnut leaves under pathogen stress.The relative expression of the two genes in the fragrance and fragrance of the two trends showed a trend of first increase and then decline,and the relative expression of the gene in the fragrance was significantly higher than the other The relative expression of.For the further study of PR1 and PR4 in the walnut in the function laid the foundation.
