Pumpkin is an important vegetable in China,and has good medicinal value in the prevention and cure of diabetes,the reduction of cholesterol,the prevention of cancer and the fight against cancer.Pumpkin is still the main means of pumpkin breeding in China.The breeding speed is slow.The establishment of pumpkin regeneration system in vitro can provide a technical basis for genetic improvement of pumpkin cell level and molecular level.Establishment of in vitro regeneration of immature embryos in this study,pumpkin pumpkin,cotyledon as material,through the cotyledons of callus and plant regeneration pathway of immature embryo callus regeneration of immature embryo-way or-way of plant regeneration,in vitro plant regeneration method.The main results are as follows:1.The best medium for inducing the callus formation of Southern Cotyledonary Nodes is: MS+1.0mg·L-16-BA+0.1mg·L-1NAA,the induction rate was 83.33%,the shortest time for the callus induction medium best 12 D.callus adventitious buds: MS+3.0mg·L-16-BA+0.2mg·L-1 NAA and B5:2.0mg·L-16-BA+0.1mg·L-1 NAA+0.5mg·L-1 GA3.for the highest rate of adventitious buds.88.89%.The best medium for callus induction is C7:MS+3.0mg·L-16-BA+0.2mg·L-1NAA,B5:2.0mg·L-1 6-BA+0.1mg·L-1 NAA+0.5mg·L-1 GA3.2.Induction of adventitious roots in cotyledon node callus: induction of adventitious roots by adding NAA to MS medium,optimum concentration of 0.1mg·L-1.The influence of different genotypes on callus induction rate was the highest,and the highest rate was reached to 91.67%.,and the callus number of Yongan No.2 was the lowest,25%.3.0.1% of the best 9min mercuric chloride disinfection disinfection effect of immature ovules.After pollination of immature ovules collected around as explants to turn green best,turning green rate is 59.38%.When the ovule was taken as an explant after pollination,the effect of turning green was the best,and the rate of greening was 59.38%.4.Pumpkin seedlings induced immature ovule culture medium: adding 10mg/L Ag NO3,ovule turn green best;the best medium of immature ovule enlarged to green: MS+10mg·L-1Ag NO3 +4.0mg·L-12,4-D+1mg·L-1NAA+0.2mg·L-1KT.5.The influence of different genotypes on callus induction rate was the highest,and the highest rate was reached by 91.67%..The callus rate of Yongan No.2 was the lowest,25.00%.
