Qira black sheep and Hetian Sheep in Xinjiang Hetian area are the main sheep species,the former has early sexual maturity,characteristics of annual estrus and intensive lambing,the latter half coarse wool sheep,wool quality is good,but the low reproductive rate,seasonal oestrus animal.It has been reported that melatonin(Mel)can affect animal reproductive traits to some extent.The aim of this study was to investigate the temporal and spatial relationships of melatonin and melatonin receptor expression in different breeds of sheep,To investigate the effect of Mel on the breeding performance of Hetian Sheep and Qira black sheep.The test collection cycle years the spring equinox,summer solstice,four solar term autumnal equinox and winter solstice day morning,noon,evening and at four time points of blood,Determination of Mel concentration at each time point by ELISA,At the same time,the ovaries and uterus tissues of Hetian Sheep and Qira black sheep were collected and extracted RNA,The expression of melatonin receptor(MTRN1A)gene in Mel and protein was studied by semi quantitative RT-PCR technique and immunohistochemical method in the period of Hetian Sheep and Qira black sheep mRNA.Results show:The Mel concentration in the blood of Qira black sheep was the lowest at noon on the day of the vernal equinox,which was significantly lower than that in the morning,evening and early morning(P<0.05),but the difference was not significant(P>0.05);There was no significant difference between the highest and lowest values of Hetian Sheep and Qira black sheep,but there was no significant difference between the four time points of(P>0.05);On the summer solstice,two kinds of sheep were the highest in the evening and lowest in the morning,but the difference was not significant(P>0.05);The equinox morning of Qira black sheep were significantly higher than the rest of the time(P<0.05)morning,noon,the difference was not significant(P>0.05),but the two was significantly lower than that in the evening(P<0.05);The total variation of Hetian Sheep was the same as that of Qira black sheep,but there was no significant difference between the four time points(P>0.05).On the day of winter solstice,the content of two kinds of sheep was significantly higher than that of other time points(P<0.05).There was no significant difference between the morning and the early morning(P>0.05),but significantly higher than that at noon(P<0.05).Year period,Qira black sheep Mel was significantly higher than the summer solstice and equinox autumnal equinox(P<0.05),was significantly lower than that of the winter solstice(P<0.05),was significantly lower than that of summer solstice equinox,winter solstice(P<0.05);At the same time,the spring equinox in the ovary of MTRN1 A in m RNA and Protein level,the expression was the highest,the summer solstice is the lowest,significantly lower than that of the winter solstice and equinox(P<0.05),and was significantly lower than that of the spring equinox(P<0.05),receptor expression is significantly lower than the lowest autumnal equinox,winter solstice(P<0.05),but not significantly.The difference with the summer solstice(P>0.05).The uterus MTRN1 A in gene and protein were the lowest expression in the summer solstice and equinox,and there wasno significant difference(P>0.05),but significantly lower than that of the spring equinox(P<0.05),while the three season was significantly lower than that of the winter solstice(P<0.05)protein expression.But Hetian Sheep is the blood content of Mel was significantly higher than that of the winter solstice summer solstice and equinox(P<0.05),the spring breeze and the difference was not significant(P>0.05),the lowest,and the difference was not significant(P>0.05),but the two was significantly lower than that of the spring equinox(P<0.05);Ovarian expression of MTRN1 A gene was significantly higher than that of the highest in the winter solstice,the summer solstice and equinox(P<0.05),and the difference was not significant(P>0.05),the lowest was significantly lower than that of the summer solstice,the spring equinox(P<0.05)the expression of MTRN1 A Protein was the lowest in summer,and the difference was not significant(P>0.05),but the two was significantly lower than that of the spring equinox(P<0.05),At the same time,the three groups were significantly lower than the winter solstice(P<0.05).The expression of MTRN1 A gene and receptor in Hetian Sheep was the same as that of Qira black sheep.The results showed that the content of Mel in the blood of two kinds of sheep had obvious circadian rhythm and seasonal rhythm,but the total content of Mel in the blood of Qira black sheep was higher than that of Hetian sheep;Similarly,the expression of MTRN1 A two gene in sheep ovarian and uterine tissue and receptor showed seasonal variation rose after the first drop,the gene expression of the equinox of Qira black sheep ovarian volume was significantly higher than that of Hetian Sheep,the contrary,no significant difference in other seasons;At the same time,there was no significant difference in the expression of mRNA in the uterus,and the expression of MTRN1 A receptor in the ovary of Qira black sheep was significantly higher than that of Hetian Sheep on the vernal equinox;There was no significant difference in the expression of the receptor in the uterus of the Qira black sheep and the rest of the season.The results showed that the Mel concentration in the blood during the annual cycle,the rhythm of MTRN1 A gene and protein expression varied with the variety and organization.This experiment through to the Qira black sheep and Mel content in blood and Hetian Sheep change,dynamic pattern of expression of MTRN1 A in ovary and uterus in mRNA and protein level,in order to provide some reference for the research on the influence of Mel on different varieties of sheep breeding function,in the actual production,Provide a scientific basis for breeding and management scale farms.
