Spring Viremia of Carp Virus(SVCV)is the pathogen of spring viremia of carp(SVC)and often causes acute hemorrhagic symptoms in various kinds of cyprinids,which induces serious aquaculture losses.The TRIM family protein was known to play an important role in many cellular processes,including potential antiviral activity,which has attracted lots of attention.In this study,a series experiments were performed to explore the mechanism of the anti-SVCV activity of TRIM47,and RNA-Seq technology were used to explore the mechanism of the interaction between SVCV and its host,hoping this study could provide some theoretical basis for more antiviral targets.1.Biological characterization analysis of the TRIM47 gene from common carp1)The sequence characterization of common carp TRIM47 was analyzed,the tissue distribution and expression characterize during its early development stages was also detected.2)SVCV infection decreased TRIM47 m RNA level significantly both in vitro and in vivo,but its expression was not affected by the virus at the protein level.3)The subcellular localization in FHM cells showed TRIM47 uniformly distributed in the cytoplasm at the form of tiny spots and partially localized in the mitochondria.4)Overexpression TRIM47 in FHM cells significantly decreased the m RNA level of SVCV-G gene and significantly increasing of IFN1 at the case of SVCV stimulation.2.Comparative transcriptome analysis of zebrafish(Danio rerio)brain and spleen infected with SVCV1)SVCV-infected zebrafish model was obtained by intraperitonealy inoculating at the water temperature of 17?.2)382 DEGs in brain and 926 DEGs in spleen were identified by RNA-Seq after SVCV infection.3)These DEGs were annotated into three ontologies(Biological process,Cellular component and Molecular function)after GO enrichment analysis.4)The KEGG pathway analysis showed that these DEGs were primarily related to Influenza A pathway and Herpes simplex infection pathway in brain and Tuberculosis and Toxoplasmosis pathways in spleen,and all of these pathways may be involved in response to pathogen invasion.5)3' and 5' AS events were significantly up-regulated in the spleen of SVCV-infected zebrafish.6)Fold changes of the candidate genes of qRT-PCR results were companied with RNA-Seq expression profiles.7)Preliminary validation was performed of the alternative splicing of RSAD2,which happened the exon skipping during its transcription...
