Chinese giant salamander(Andrias davidianus),also known as baby fish,belonging to Amphibian,order Caudata,family Cryptobranchidae,is the largest extant amphibian species in the world.Chinese giant salamander crowned as living fossil,is classified as a class II protected species in China.In recent years,Chinese giant salamander has retreated to a narrow strip along the enormous destroyed resources influced by multiple factors,therefore it's of great urgency to protect the Chinese giant salamander.Chinese giant salamander belonging to amphibians representing the transition from aquatic to terrestrial life with poor migration and prey ability,is an ideal material for study of evolution and biodiversity.The small intestine of Chinese giant salamander is an important organs of digestion and absorption.In ultrastructure,small intestine of Chinese giant salamander already has the structure for strong digestion,absorption,synthesis and secretion,for example,rich mitochondria,endoplasmic reticulum,golgi bodies and lysosomes has been observed in cytoplasm.At present,the study of Chinese giant salamander mainly focused on histomorphology,research have little been reported at the molecular level.In our study,we mainly adopted advanced i TRAQ coupled with 2D LC-MS/MS technology for proteomics,to test the ptoteins in the small intestine of Chinese giant salamander.Then,these ptroteins were classified as physiological activities by bioinformatics methods(e.g.GO,NCBI,GENMAPP,KEGG,QIAGEN and IPA).Subsequently,the relevant proteins participating in carbohydrate metabolism,lipid metabolism,amino acid and protein metabolism,and nucleotide metabolism,were selected out,and were conducted pathway analysis by IPA.Finally,the funcion of major proteins in the relevant pathways were analyzed by consulting relevant literatures.Main results of the study are as follows:(1)117 proteins took part in carbohydrate metabolism in the small intestine of Chinese giant salamander.The major physiological activities are glycolysis and gluconeogenesis with the major ptoteins ALOA,ALOB,ENO1,ENO2,ENO3,GPI and so on,and pentose phosphate pathway with major ptoteins PGD,PGLS and so on.(2)142 proteins took part in lipid metabolism in the small intestine of Chinese giant salamander.The major physiological activities are fatty acid ?-oxidation?with major ptoteins ACADM,ACSL1,ECHS1,HADHA,HADHB and so on,and ketogenesis with major ptoteins BDH1,HMGCS1 and so on.The AMPK Signaling were detected in both carbohydrate metabolism and lipid metabolism with major proteins CYP7B1.(3)121 proteins took part in amino acid and protein metabolism in the small intestine of Chinese giant salamander.The major physiological activities are protein ubiquitination pathway with the major ptoteins PSMC1,PSMC2,PSMC3,PSMC4,PSMC5 and so on,and histamine degradation with major ptoteins DAO and so on.The major signaling pathway is AHR signaling pathway with the major ptoteins SLC7A8,SLC3A1 and so on.(4)105 proteins took part in nucleotide metabolism in the small intestine of Chinese giant salamander.The major physiological activity are Purine nucleoties De novo Biosynthesis?with the major ptoteins AK1,NME7,NUDT9,UMPS and so on.The major signaling pathway is SAPK/JNK signaling pathway with the major ptoteins CDC42,CRK,CRKL and so on.In conclusion,our study used iTRAQ coupled with 2D LC-MS/MS technology to detect the expression abundance of proteins participating in carbohydrate metabolism,lipid metabolism,amino acid and protein metabolism,and nucleotide metabolism in the small intestine of Chinese giant salamander,and the methods of bioinformatics to analyse the physiological activities and relevant signaling pathways happening in the small intestine ofChinese giant salamander,and the method of consulting relative references to explore the funcion of the related proteins joining in adjusting metabolism,which provided clues to furhter investigate the physiological activity of the small intestine of Chinese giant salamander.
