In this study,mammary epithelial cell lines(EpH4-Ev and MAC-T)and bacterial(Escherichia coli NJ0501,E.coli NJ0501 and Staphylococcus aureus WX0801,S.aureus WX0801)were chose as the research objects.Tlhen,the cells were challenged with E.coli NJ0501 or S.aureus WX0801 to investigate the innate immune response of different cells;and to reveal the effect of taurine on these inmune response.The results were listed as follows:1 Comparison the response of mouse and bovine mammary epithelial cells to the infection of bacterialTo investigate whether EpH4-Ev cells can be used to substitute MAC-T cells in the study of cells innate immune response.Cells were challenged with bacterial and the NAGase activity in cells supermatant and levels of ROS in cells were detected.The results showed that the infection of both pathogens can cause similar inflammation in these two mammary epithelial cells,and the pretreatment of taurine can reduce pathogens induced inflammation in both cells.Moreover,the activity of NAGase in cell supernatant and levels of ROS in cells significantly increased in both mammary epithelial cells(P<0.05).The results showed that EpH4-Ev cells can be used to substitute MAC-T cells as challenge cells.2 Escherichia coli-induced innate immune response in mammary epithelial cells and the regulation of taurineIn order to investigate the cell inflammation changes and the role of taurine on cells innate immune response after mammary epithelial cells challenged with E.coli NJ0501.Mammary epithelial cells and E.coli NJ0501 were chosen as the research objects.We first detected the cell inflammatory changes after the incubation of mammary epithelial cells with E.coli NJ0501 and further explored the protective effect of taurine on cell inflammatory injury.Experimental results showed that two hours after the infection of mammary epithelial cells with E.coli NJ0501,the expression of mRNA(TLR2,TLR4,MyD88,iNOS,TNF-?,IL-1?),protein(TLR2,TLR4,MyD88),the activity of NAGase in cells culture supernatants,and the levels of ROS and MDA in cells increased significantly(P<0.05).In addition,the activity of SOD and the expression levels of T-AOC decreased significantly(P<0.05).However,the pretreatment of mammary epithelial cells with taurine significantly reduced the expression levels of mRNAs(TLR2,TLR4,MyD88,iNOS,TNF-?,IL-1?,P<0.05)and proteins(TLR2,TLR4,MyD88,P<0.05).Moreover,taurine significantly decreased the activity of NAGase,the levels of ROS and MDA(P<0.05),and increased the activity of SOD and T-AOC(P<0.05).The results showed that taurine could attenuate the inflammation of mamnary epithelial cells induced by E.coli NJ0501.3 Staphylococcus aureus-induced innate immune response in mammary epithelial cells and the regulation of taurineTo elucidate the cell inflamnation changes and the role of taurine attenuation on innate immune response after mammary epithelial cells challenged with S.aureus WX0801.Mammary epithelial cells and 5.aureus WX0801 were chosen as the research objects.The cell inflammatory changes after incubation of mammary epithelial cells with S.aureus WX0801 were detected and further explored the protective effect of taurine on cell inflammatory injury.Experimental results showed that two hours after the infection of mammary epithelial cells with S.aureus WX0801,the expression levels of mRNA(TLR2,TLR4,MyD88,iNOS,TNF-?)and protein(TLR2)and the activity of NAGase in cells culture supermatants,the levels of ROS and MDA in cells increased significantly(P<0.05).In addition,the activity of SOD and T-AOC decreased sigificantly(P<0.05).The pretreatment of mammary epithelial cells with taurine significantly reduced the expression levels of mRNA(TLR2,TLR4,MyD88,iNOS,TNF-?,P<0.05)and protein(TLR2,P<0.05),the activity of NAGase,the levels of ROS and MDA(P<0.05),and increased the activity of SOD and T-AOC(P<0.05).The results showed that taurine could attenuate the inflammation changes of mamnary epithelial cells induced by the S.aureus WX0801.
