Nuclear pore proteins are integral components of the nuclear pore complex,which mediate the nuclear cytoplasmic transport of various macromolecules,including regulators of plant defense responses.Signaling of plant defense responses and nuclear cytoplasmic transport are inseparable,so the nuclear pore proteins play an important role in plant defense responses.Aquaporins constitute a biofilm channel mediate intracellular material transport,such as H2O2,and participate in a variety of physiological and pathological reactions of plants.H2O2 is a kind of reactive oxygen species,impacts on plant disease resistance.The issue is for the nuclear pore protein At1G13120 and aquaporin AtPIP1;4,in order to understand the plant resistance immune mechanism.The main content of this paper is divided into two parts:The first part:Screening resistance of the nine nuclear pore mutants,get two kinds of mutants which can mediate plant defense resistance.Take Salk120410 as the research object,nuclear pore protein gene At1G13120 for study,we found that the gene can be induced expression by SA and mutating At1G13120 can significantly inhibit the expression of resistance-related genes PR1 and PR2,indicating that At1G13120 involved in the SA signaling pathway mediated defense responses.On the other hand,mutating this gene can reduce their resistance to Pst DC3000,but can promote growth and increase the amount of seed production.The second part:The obtained AtPIP1;4 mutants,the wild-type and transgenic plants was measured and compared for the resistance.The results showed that AtPIP1;4 transgenic plants more resistant than the wild-type and mutant plants are more susceptible than the wild type.Experiments confirmed that AtPIP1;4 can regulate the expression of PR genes,and participate in SA signaling pathways.To further explore the relationship between AtPIP1;4 and H2O2,found that AtPIPl;4 had not directly induce the accumulation of hydrogen peroxide in leaves,but can assist pathogen-induced apoplastic hydrogen peroxide metastasizing into the cytoplasm.To sum up,changing the expression of Arabidopsis nuclear pore protein gene At1G13120 or aquaporin gene AtPIP1;4 can lead to the alter of Arabidopsis sensitivity to Pst DC3000,both of which can regulate the expression of disease-related genes of SA signaling pathways,thus have an impact on plant disease resistance.
