Strawberry is a polyploid plant,and the polyploidy(2x,4x,6x,8x,10x)strawberrys of germplasm resources are widespread in nature.Wild strawberry is an important gene pool for the disease resistance,stress resistance and improvement of strawberry.It is often used to create new germplasm by the cross breeding of cultivated strawberry and wild strawberry.In my study,pentaploid strawberry(code name:FxLs-11-37,2n=5x=35),which was derived from the hybridization of diploid green strawberry(Fragaria viridis Duch.)and octaploid strawberry(Fragaria x ananassa 'Fusanoka'),and its autodecaploid(code name:A3,2n=10x=70)were employed for conducting the proteomic analysis with two dimensional gel electrophoresis technology and iTRAQ technology,we aimed to obtain the key genes that control important characteristics of strawberry during chromosome doubling.The main results were as follows:1.Pentaploid strawberry(FxLs-11-37,2n=5x=35)derived from the hybridization between diploid Fragaria viridis Duch and octaploid F.x ananassa 'Fusanoka',and its autodecaploid(A3,2n=10x=70)were employed for conducting proteomic analysis with two dimensional gel electrophoresis technology.Gel pictures of good resolution and repeatability were obtained.Determination of strawberries agronomic traits,SPAD value and the net photosynthetic rate,Compared with FxLs-11-37,A3 showed a significant dwarf type,larger crown,increased leaf length and leaf width,much thicker in leaf,more dark green leaves,and it also has a bigger SPAD for leaf and net photosynthetic rate;Protein spots of FxLs-11-37 and A3 analyzed using PDQuest software were mainly scattered in the isoelectric point ranging from 4 to 7,and molecular weight ranging from 14.4 to 66.2kDa.About 700 expressed protein points in FxLs-11-37 and A3 were detected.Expression levels of 18 proteins changed over 1.5 fold after chromosome doubling and 4 changed over 2.0 fold.We identified these 4 proteins with MALDI-TOF-MS/MS mass spectrometry technology,and results showed that the 4 proteins are major strawberry allergen Fra a 1-E,heterogeneous nuclear ribonucleoprotein 1,lipoyl synthase 1,chloroplastic,and NAD(P)H dehydrogenase(quinine)FQRl-like.We also studied the transcription difference of 4 genes encoding the 4 differentially expressed proteins.Results showed that there were positive relations between the transcription levels and proteins contents.Analysis indicated that these proteins are involved in stress resistant,mRNA transport,material and energy metabolism.2.In this study,we used pentaploid strawberry(code name:HjLs-11-09),which derived from the hybridization of Fragaria viridis Duch and octaploid strawberry(F.×ananassa 'Benihoppe'),and decaploid strawberry(code name:D-H-09-01 2n=10x=70,HjLs-11-09 chromosome doubling)as materials to conduct the proteomic analysis with iTRAQ technology.The expressed abundance of 118 protein proteins spots had 1.5-fold changes after chromosome doubling,and the abundance of 106 was increased after chromosome doubling.The expressed abundance of 15 protein proteins spots had 2.0-fold changes after chromosome doubling,and the abundance of 13 was increased after chromosome doubling.We identified these differentially expressed proteins,and made a GO function analysis.Most of them are mainly involved in the proteolysis,lipid transport,DNA dependent transcription,mRNA processing,protein folding,nuclear mRNA splicing,nucleosome assembly,and cell division processes for biological process;Most of them are mainly located in in the nucleus,cytoplasm,organelles and cytoplasmic region,the plasma membrane and integrated membrane protein for cellular component;Most of them are mainly participating in enzyme catalytic activity and binding function for molecular function.We also conducted the KEGG analysis for the differentially expressed proteins,We also studied the transcription difference of 4 genes encoding the 4 differentially expressed proteins.Results showed that there were positive relations between the transcription levels and proteins contents.and found that most of them are involved in carbohydrate metabolism,genetic information,signal transduction,energy metabolic and other pathways.And chlorophyll metabolic pathway is the main factor.Chlorophyll metabolism related proteins:cytochrome P45081D1,pheophorbide a oxygenase,chloroplastic,29kDa ribonucleoprotein A,chloroplastic;Signal transduction related proteins:rac GTP-binding protein 5,elicitor-responsive protein 3;Secondary metabolism related proteins:GDSL esterase/lipase At5g03810;the stress response and defense-related proteins:heat shock cognate 70 kDa protein.
