Straw as a kind of renewable resources is extremely rich on the earth,but the problems is that production is big,traffic inconvenience,serious waste of resources and environmental pollution result from most of the straw is burned or discarded.In order to solve the problem of slow speed of straw in natural conditions,and with the aim of accelerating the decomposition of straw to make organic fertilizer to provide resources and technical support,18 cellulose-decomposing strains were isolated and screened from different types of soils rich in litter,which consisted of 12 actinomycetes and 6 bacteria.3 high cellulose degrading strains(Z1?Z2?Z13)including 2 actinomycetes(Z1?Z2)and 1 bacteria(Z13)were selected by compound sieve measuring the diameter of the hydrolysis circle to judge the ability of cellulose.The results of identification through physiological and biochemical test,culture characteristics,and 16S rRNA sequencing analysis showed that Zl and Z2 belonged to genus of Streptomyces,Z13 was genus of Bacillus cereus.The results of decomposing filter paper and cellulose enzyme activity among each single bacteria and the mixed strain showed that each strain had certain capability of decomposing cellulose,but mixed strain behaved the best and two strains bacteria had obviously synergistic effect,Z1?Z2?Z13 and the mixture were started to decompose filter paper at 64 h?60 h?60 h 50 h,and the mixture could completely decompose the filter paper at 96 h,effects are better than each single strain.The highest enzyme activity of Z1?Z2?Z13 were 32.08 IU/mL,32.08 IU/mL,7.8 IU/mL and the enzyme production peak comes at 9th day,8th day,3th day respectively,actinomycetes is in the intermediate level,and bacterial Z13 above the level of the current research has been reported.The best conditions of cellulose secreted by Z13 showed that the optimal pH value was 6,the optimum temperature was 50 ?,and the specific activity was 232.1 IU/?g under the condition of suitable for high temperature stage of composting.And both of the cellulase synthetized by three bacteria can be secreted to the outside of the cell,there are great development prospects in the application of cellulose degradation.After liquid fermentation and solid state fermentation,has been produced compound rotten agent on three bacteria.The experiment were set up two treatments for aerobic compost text with crop straw including no-inoculant(CK),self-inoculantion of microbial group.And 5 indicators were traked by temperature,pH,water content,tension and the numbers of microorganism,the result showed that experimental fast into the high temperature period and can maintain 15 days,30 days has been decomposed completely,while the control group in 40 days has not been completely finished.Indicate that access to the compound rotten agent can obviously accelerate organic matter decomposition rate,speed up the process of rotten,so as to provide theoretical basis for the future straw returned and technical support.
