The study was conducted to investigate the influence of fatty liver hemorrhagic syndrome on varies from oxidative stress and inflammation perspectives,and also study the regulatory mechanism of resveratrol.A total of 144 Hyline brown laying hens aged 90 days were randomly divided into 4 groups with 36 birds in each rgroup and were fed with the different of diets.Control group was fed with basic diets;HELP group was pathologic model group with birds fed with high-energy-low-protein diets;Con+Res group and HELP+Res group were fed with basic diets and high-energy-low-protein diets containing a resveratrol dosage of 400 mg/kg,respectively.The study lasted for 120 days,and the production data was recorded every day.Ovary samples were collected on days 40,80 and120 to determine the content of malonaldehyde(MDA)and the activities of catalase(CAT),superoxide dismutase(SOD)and glutathione(GSH);as well as the messenger RNA(mRNA)expression levels of anti-inflammatory related genes(NF-?B?TNF-??IL-1??IL-6)and antioxidant-related genes(Nrf2?HO-1?SOD-1);and also determine the expression of FSHR and LHR.The results were as follows.1.The level of laying rate was significantly declined(P<0.01)in HELP group compared to control group,while it was significantly increased(P<0.01)in HELP+Res group.2.The content of MDA was significantly increased(P<0.05 or P<0.01)and the activities of SOD,CAT,GSH were significantly declined(P<0.05 or P<0.01)in HELP group compared to control group.While the contents of MDA were significantly(P<0.05 or P<0.01)declined,and the activities of SOD,CAT and GSH were significantly(P<0.05 or P<0.01)increased in HELP+Res group.3.The mRNA expression levels of NF-?B,TNF-?,IL-1? and IL-6 were significantly(P<0.05)increased in HELP group compared to control group while significantly(P<0.05)decreased in HELP+Res group.4.The m RNA expression levels of Nrf2,HO-1 and SOD-1 were increased(P<0.05)in HELP and significantly increased(P<0.01)in HELP+Res group compared to control group.5.The mRNA expression levels of FSHR and LHR were decreased(P<0.05)in HELP and significantly increased(P<0.01)in HELP+Res group compared to control group.In conclusion,FLHS caused an increased level of lipid peroxidation in ovaries which leaded to the oxidative stress and furthermore activated the NF-?B signaling pathway in response to inflammation reaction,finally resulting in ovariy dysfunction and decline of egg production.However,resveratrol effectively ameliorated oxidative stress and inflammation in ovaries of FLHS hens,Which might be associated with its role in the activation of Nrf2 signaling pathway and inhibition of NF-?B signaling pathway.
