Development Of The Colloidal Gold Immunochroatographic Rapid Test Strip For The Determination Of Veterinary Drug And Illegal Additions

In this study, it developed the detection of norfloxacin, sulfaquinoxaline and melamine residue based on the colloidal gold immunochromatograph assay (GICA) and successful-ly used this method to detect the animal food products.Developing norfloxacin colloidal gold immunechromatography test strip detection method, through the optimization, the best work condition is: the particle size of colloidal gold which is prepared by the method of trisodium citrate reduction is 18 nm to labeled antibody; the optimal pH value that gold articles labeled antibody is 9.0; the suitable quantity of antibody is 18 ?g/mL; the detection time is less than 10 min; the limit detection is 2 ng/ml. There is cross-reaction with other quinolones, but no cross-reaction with other kinds of veterinary drugs which means the method has some specificity. The limit detection in liver, pork, beef, chicken breast, sea bass, samples is 20 ng/g. The limit detection in milk is 12 ng/mL.Through the optimization, the best work condition of the detection of sulfaquinoxaline by GICA is: the particle size of colloidal gold is 18 nm, the optimal pH value that gold articles labeled antibody is 8.5; the suitable quantity of antibody is 24 ?g/mL; the detection time is less than 10 min; the limit detection is 10 ng/ml. There is cross-reaction with sulfa-chloropyridazine (SCPA), sulfamethoxazole (SPMX), sulfamethoxazole(SMT), Sulfaquinoxaline (SQ) which are belong to the same kind, sulfonamide, but no cross-reaction with other kinds of veterinary drugs and sulfonamide which means the method has good specificity. The limit detection in liver, pork, beef, chicken breast,shrimp, egg samples is 80 ng/g.Through the optimization, the best work condition of the detection of melamine by GICA is: the particle size of colloidal gold is 18 nm, the optimal pH value that gold articles labeled antibody is 8.0; the suitable quantity of antibody is 24 ?g/mL; the detection time is less than 10 min; the limit detection is 500 ng/mL. There is no cross-reaction with triazine structural analogues of cyanuric acid, atrazine, cyanuric chloride but cyclopropylamine which means it has good specitificity. It is not necessary for milk samples to be treated before being detected. The limit detection of milk is 500 ng/mL.
